Biofortification of mushroom (Pleurotus floridanus) using calcium based supplements

The nutritional value and yield of mushrooms depend on the substrate on which it is grown. This study sought to biofortify Pleurotus floridanus with calcium supplements and assess its effect on the yield and calcium levels. The experiment was set up in a 2 × 5 factorial and replicated thrice in a completely randomized design. Two calcium supplements, OML and OMW, were added to two growth media. The examination of total dry weight yield showed that calcium supplements OML and OMW in the sawdust medium containing wheatbran in the ratio 1:10 had a mean value of 4.37 g, which was significantly higher (P < 0.05) than that in the control (1.29 g). However, in the sawdust-only medium, there was no significant difference (p > 0.05) in the application of treatments. No significant difference (p > 0.05) was observed between the calcium types in both growth media. The mineral analysis showed that calcium levels were increased in harvested mushrooms with the addition of calcium OML and OMW to the growth media.

This study surveyed mushrooms as part of the Hallasan Natural Reserve Basic Academic Survey, which was conducted for the purposes of securing basic data on the Hallasan Mountain Natural Reserve; our study targeted the area between Seongpanak Trail and Donnaeko Trail from April to October 2018. A total of 392 species from 184 genera in 65 families were investigated. Among them, 187 mushrooms were wood-decaying species. The greatest diversity in mushroom species was recorded in July (206 species) at an altitude of 600-800 m above sea level. Among the mushrooms investigated, five were climate-sensitive biological indicator species and two were potential candidates for that classification. Of these, 163 species were biological resources that require approval for overseas export.

Five varieties of mushrooms, including Sae-do, were examined using farm field tests to determine their morphological characteristics and quality by performing harvest flush. Considering the cultivation characteristics of the varieties studied, the parts related to mushroom generation and production showed almost similar characteristics. Overall, the mycelial forces were similar; however, in the evaluation of plantation tests performed by farmers, "saedo" was found to be the strongest and "saehan" was considered to be the weakest. Weights of fruiting bodies were related more for compost status and regimen of use than for breeding, and it was difficult to distinguish between the weights of fruiting bodies of the varieties studied. Regardless of the variety, pileus diameter, pileus thickness, and stem length of Dalseong-gun, Yongin-si, and Boryeong-si decreased as the harvest flush progressed. However, stem thickness tended to increase in the second and third flush compared to that in the first flush, regardless of the variety. However, the Gyeongju-si farms could not identify any trends attributable to varieties and harvest flushes. The pileus size and stem length of the fruiting bodies of varieties from Buyeo farms decreased with the progression of the harvest flush. However, pileus thickness and stem thickness did not show any specific tendency of growth among the varieties studied. The differences in the morphological characteristics of mushroom varieties in each harvest cycle are expected to be caused by differences in the supply of nutrients in the medium, the quality of the compost used, and the cultivation environment rather than the variety.

The cultivation of Sparassis crispa in the beginning of the 2000s in South Korea, and the cultivar ‘Neowul’ bred in the Chonbuk Agricultural Research and Extension Service were registered first in 2016. However, there is no manual for the cultivation of Sparassis crispa, and therefore, there remains a big difference in its harvest rate across farms. Herein, we aimed to study the primordium formation conditions of Sparassis crispa ‘Neowul’ according to the medium pH, medium moisture content, and inoculum volume of liquid spawn and develop a stable production technology. We found the annual yield per bottle relating to the cultivation period, harvest rate, and the weight of fruiting body to be the highest at 363.6 g in the area cultivated at pH 3.8. However, it is thought that cultivation by adjusting the pH to 3.9±1 would be necessary for stable production, considering that at pH 3.6, the yield sharply reduced to 189.5 g. Moreover, the culture period was shorter at pH 4.0 compared with that at pH3.8, and the cultivation period at pH 4.0 was the same as that at pH 3.8. No significant difference in the weight of the fruiting body at different conditions was recognized. Additionally, it is difficult to regulate the pH precisely in practical applications in the farms. It is thought that 341.8 g Sparassis crispa will be produced per bottle annually if the medium moisture content is adjusted to 65%, liquid spawn inoculum volume is equivalent to 4% of the medium volume, and the humidity in the culture room is set to below 50%.

The effect of the nitrogen content of sawdust medium (1.2~1.8%) and the restriction stage temperature (2, 4, and 6oC) on the growth and storage characteristics of Flammulina velutipes (winter mushroom) were investigated. With increased nitrogen content, the growth days shortened and the yield of the fruiting body increased. The effect of restriction temperature on the growth of the fruiting body differed depending on the nitrogen content. No difference in restriction temperature was evident for T1 with a low nitrogen content of 1.28%. In medium with a nitrogen content ≥1.55%, the yield and length of the pileus and stipe increased as the restriction temperature decreased. The weight loss ratio according to the storage period did not change according to the nitrogen content in the medium. A low weight loss ratio of 1.50 to 1.93% was observed at a restriction temperature <4oC. When T3 with high nitrogen content in the medium was treated at a restriction temperature of 4°C, the fruiting body color values after 31 days of storage were 84.81 (L) and 6.3 (ΔE). This color change was minute compared to that after other treatments. The sensory characteristics score was 5.2 after 31 days of storage, and the quality remained acceptable for consumption.

This experiment was conducted to solve the failure of fruiting body production in the bottle cultivation of the oyster mushroom cultivar ‘Heuktari’. The effects of incubation temperature on primordium formation and fruiting body yield of the oyster mushroom cultivar ‘Heuktari’ were investigated. The proper temperature for mycelium growth of ‘Heuktari’ on potato dextrose agar (PDA) medium is 23–26oC. The mycelial growth of ‘Heuktari’ was faster than that of Chunchu 2ho. During mycelial culture in sawdust medium, the temperature of the medium in the bottle initially increased, reached the highest point in the middle of the culture, and then decreased. The higher the set temperature, the shorter the incubation period. When the incubation temperatures were 20°C and 24°C, respectively, the undeveloped primordium formation rates were low (1.8% and 4.2%, respectively). However, the rate of undeveloped primordium formation increased, and the yield decreased at incubation temperatures of 16°C and 28°C. Mushroom farms that set incubation temperatures to 18°C and maintained the medium temperature at less than 28°C showed undeveloped primordium formation rates ranging between 0.3-0.8%. The rate of undeveloped primordium formation increased and the yield decreased in the farms with high incubation temperatures (above 28°C). We found that in order to reduce undeveloped primordium formation, the air inside the incubation room should be circulated continuously so that the temperature of the medium does not rise above 28°C, and dense incubation conditions should be avoided.

Agaricus bisporus is an important edible mushroom that is used as a functional food. In this study, European A. bisporus strains were analyzed for genetic diversity, population structure, and genetic differentiation using simple sequence repeat (SSR) markers. European A. bisporus strains were divided into four groups by distance-based analysis and two subpopulations by model-based analysis. The SSR markers used in this study did not group European A. bisporus strains by geographical region or pileus color. Genetic diversity was high in Group 4 based on distance-based analysis and Pop. 2 based on model-based analysis. A. bisporus strains showed very low genetic differentiation. The results of this study can be used for breeding A. bisporus in the future.

This study was conducted to cultivate new Lepista nuda varieties with shorter cultivation period and better fruiting body compared to that of wild strains, for mass production and commercial application. Eighteen genetic resources of L. nuda were collected and grown in boxes using rice straw-fermented growth medium. Four lines with fruiting bodies were formed and selected as cross-breeding lines. Although 657 combinations were crossed through monospore crossing, only 17 combinations were bred between the ‘CBMLN-19’ line and the ‘CBMLN-30’ line. Among them, 8 lines with fast mycelial growth and high density were selected. After inoculating the rice straw-fermented growth medium with 14 genetic resources and 8 cross-breeding lines, their incubation period was investigated. Six of the cross-breeding lines completed their incubation in 20 days, while 7 of the 14 genetic resources took more than 40 days to complete their incubation, reducing the incubation period by more than 20 days in most cross-breeding lines. After the incubations were completed, the clay loam soil was covered with for post-cultivation, and when the mycelial cultivation was complete, the formation of fruiting bodies was induced after scraping the mycelial bodies under these environmental conditions: 14oC, 95% relative humidity or higher, and 1,500 to 2,000 ppm CO2 concentration. The temperature was reduced to 6oC at night, resulting in a low temperature shock. Thus, 4 lines of fruiting bodies occurred from two genetic resources ‘CBMLN-31’ and ‘CBMLN-44’ and two cross-bred lines ‘CBMLN-96’ and ‘CBMLN-103’. After inoculation, the longest period for fruiting bodies to occur was 100 days for the control:, the genetic resource ‘CBMLN-31’, and the shortest period (45 days) was observed for the cross-breeding line ‘CBMLN-103’. The result of the investigation of the fruiting body characteristics shows that the cross-bred line ‘CBMLN-103’ showed a small form with 1.9 g of individual weight and 123validstipes per box, which was the highest incidence among the four lines. Another cross-bred line, ‘CBMLN-96’, had an individual weight of 5.5 g, which is larger than that of ‘CBMLN-103’; however, the number of valid stipes per box was 30 less than that of ‘CBMLN-103’. Quantity analysis showed that the control, ‘CBMLN-31’, had the highest quantity of 783 g per box, followed by the cross-bred line, ‘CBMLN-96’ with 165 g per box, and then the ‘CBMLN-103’ with 232 g. The quantity of the two crossbred lines was lower than that of the control ‘CBMLN-31’; however, the amount of fruiting bodies was higher, and the cultivation period was shortened by 32 to 33 days. Therefore, these two lines would be selected as superior lines.

This study aimed to optimize total nitrogen (TN) content in the medium and the growth temperature to improve the shelf life of Pleurotus eryngii Keneutari No. 2. We investigated the growth characteristics and changes in the O2/CO2 content in polypropylene film and performed sensory evaluations during cold storage under 1.5%, 1.8%, 2.1%, and 2.4% TN and at temperatures of 9°C, 12°C, and 15°C. The diameter and weight of the fruit bodies tended to increase with increasing nitrogen content, regardless of the growth temperature, and the highest weight per bottle (149.5 g) was obtained at 12oC and 2.4% TN. Regardless of the nitrogen content, leaf color deepened with decreasing temperature. The gas concentration in the film, which changes depending on storage time, was maintained for the longest period at 9oC and 2.1% TN. The sensory evaluation revealed no difference between the different nitrogen conditions tested, and the odor, discoloration, and decay were similar at 15oC and 9oC until day 28 of storage.

This study aimed to investigate the biological activities and amino acid content in the mixtures of roasted edible mushrooms (Pleurotus ostreatus, and Lentinula edodes) and grain additives (oat and brown rice) obtained under different extraction conditions for the development of mushroom tea. The total amount of polyphenol and β-glucan in edible mushrooms was increased with the roasting treatment compared to that observed with the air-drying treatment. In addition, DPPH radical scavenging activity and the amount of all amino acid components, including glutamic acid (Glu), were higher in the oat extracts than those observed in the brown rice. The biological activities and nutrient content were examined based on extraction temperatures and thetime required to obtain the mixtures of edible mushrooms and grain additives. The extract of a 1:1 mixture of L. edodes + oat (LE+O) obtained at 100oC, 3 min showed high DPPH radical scavenging activity (33.5%), and the extract of 3:1 mixture of P. ostreatus + brown rice (PO+B) obtained at 100oC, 3 min showed high nitrite scavenging activity (49.9%). The total polyphenol content of the extract of 3:1 mixture of P. ostreatus + brown rice (PO+B) was the highest when obtained at 16.2 mg GAE/g and+ brown rice (PO+B) was the highest when obtained at 16.2 mg GAE/g and 70oC, 10 min . The essential amino acid content was higher in the 1:1 mixing ratio of P. ostreatus + oat (PO+O) and P. ostreatus + brown rice (PO+B) and 3:1 mixing ratio. L. edodes + oat (LE+O) and L. edodes + brown rice (LE+B) compared to that observed in other mixing ratios.

The grade and price of Lentinula edodes largely differs in preference depending on the product area and seasonal factors. The product amount of autumn L. edodes was higher than that of spring L. edodes, but high quality, which is divided into “Hwago” is low in preference. Mostly, the autumn L. edodes is obtained as powder; hence, it is necessary to develop a processing method to utilize its flavor and aroma at an affordable price. Additionally, we estimated the content of β-glucan, ergosterol, vitamin D2, reducing sugars, and free amino acids and evaluated the anti-inflammatory activity of saccharification powder of log-cultivated L. edodes. In the saccharification powders obtained via 7 min of UV irradiation of log-cultivated L. edodes, β-glucan and vitamin D2 contents were found to be the highest, whereas ergosterol content was found to be the lowest. The content of reducing sugars ranged from 62.4 mg/L to 68.2 mg/L. The free amino acids were higher in these saccharification powders than in the control. Subsequently, RAW 264.7 cells were treated with different concentrations (10, 50, 100, 200, 300, and 500 μg/mL) of the saccharification powders of log-cultivated L. edodes obtained via different UV irradiation time applications. The cells showed good viability; the antiinflammatory effect was found to be the highest at 7 min UV irradiation. Therefore, 7 min of UV irradiation was determined to be the optimum condition for manufacturing saccharification powders of log-cultivated L. edodes. Hence, saccharification powders of log-cultivated L. edodes may be used as a raw material for natural sweeteners, food additives, and in the food industry.

In this study, we analyzed the effect of media on the production of carotenoids and mycelia by using Perenniporia fraxinea. Malt extract-based medium with less than 0.1% peptone stimulated the production of carotenoids, and the one with more than 0.2% peptone inhibited its production. P. fraxinea grown in medium without malt extract did not produce carotenoids, although a small amount of peptone was added to the medium.After carotenoid production, the culture broth was separated using simple centrifugation and the supernatant was harvested as a carotenoid solution. Ethanol was used to extract carotenoids from mycelia. Carotenoid solution separated or extracted from the culture solution showed DPPH radical scavenging activity. The antioxidant carotenoids produced by P. fraxinea are derived from natural products, have no toxicity and side effects, and exhibit excellent antioxidant effects; therefore, they can be effectively used to remove oxides produced by active oxygen.

Snow fungus (Tremella fuciformis) with functional contents has satisfactory effects on various diseases, including atherosclerosis, high cholesterol, healthy skin, cancer, diabetes, and anti-inflammation. In this study, the extraction yield and functional contents (ergothioneine and β-glucan) of white jelly fungus were compared based on the extraction conditions using response surface methodology. Results revealed the extraction conditions for optimization of the dependent variables to be 60oC and 4.33 h, when 16.6 mg/mL of sample concentration was used. Under these conditions, the extraction yield was 24.9%, including ergothioneine (66.8 μg/g) and β-glucan (34.9 g/100 g). These results can be useful in understanding the functional ingredients and mass extraction process in mushroom.

The study was conducted for the safety evaluation of 320 pesticide residues in 768 Lentinula edodes fruit body samples and 143 L. edodes media samples, which are distributed nationwide in South Korea. The monitoring method was the second of the multi-residue methods in the Korean Food Code. GC-ECD, GC-NPD, and GC-MSD were used as evaluation equipment for analysis. Single-analysis of the target pesticides was performed for mepiquat chloride. Through the analysis of collected L. edodes samples, pesticide residues were detected in total seven cases, including four L. edodes fruit body samples and three L. edodes media samples. The detected pesticide residues were carbendazim, diflubenzuron, fluopyram, and dinotefuran. In this study, carbendazim was detected in three L. edodes fruit body samples and one L. edodes media sample. The detected amount of carbendazim was 0.056, 0.17, 0.043, and 0.09 mg/kg, respectively. The amount of carbendazim in the collected L. edodes samples was detected below the MRLs (maximum residue level). The detected amounts of fluopyram and dinotefuran were 0.068 mg/kg and 0.06 mg/kg, respectively. Two pesticide residues were detected in the medium in one case. Mepiquat chloride was not detected in this study. These results suggested that residual pesticides were detected in a small number of collected L. edodes. However, the PLS for unregistered pesticides MRL was 0.01 ppm; therefore, we have to conduct research on preparing safety standards for mushrooms, including L. edodes.

Eight dried ear mushroom products (three domestic and five Chinese products) distributed in Korea were analyzed for 321 residual pesticides, 7 heavy metals, and 3 types of radioactivity. Residual pesticides in the domestic products were not detected. However, chlorpyrifos, isoprocarb, mepiquat chloride, and carbendazim were detected in four Chinese products, all of which were below the allowable residual limit. Among the detected pesticide ingredients, only the residue for mepiquat chloride has been established to be present in ear mushrooms. In the heavy metal test, trace amounts of heavy metals were detected in all samples. However, none of the samples exceeded the allowable residual limits, except for one domestic sample that exceeded the standard value of 0.3 mg/kg for lead. Radioactivity tests confirmed that levels were below the minimal detectable activity value in all samples.

Mycelium composites and leathers have versatile material properties based on their composition and manufacturing process. To prepare mycelium mat for the production of mushroom leather, several strains were mutated by gamma rays. Some mutant strains, including Lentinula edodes, Ganoderma lucidium, and Schizophyllum commune showed good hyphae growth rate and density on saw-dust media. Irradiation power (Gy), time, and height from the radiation source to the sample were examined. Based on the preliminary data obtained in this study, comprehensive research should be conducted to explore the optimal strains and culture conditions for mycelium-based leather production.

This study was conducted to set up a proper replacement cycle of High Efficiency Particulate Air (HEPA) filters by observing the microbial populations in the air of the cultivation house of Pleurotus eryngii, before and after HEPA filter replacement at different periods . The density of bacteria and fungi in the air during each cultivation stage was measured using a sampler before the replacement of the HEPA filter. The results showed that airborne microorganisms had the highest density in the mushroom medium preparation room, with 169.7 CFU/m3 of bacteria and 570 CFU/m3 of fungi, and the removed old spaun had 126.3 CFU/m3 of bacteria and 560 CFU/m3 of fungi. The density of bacteria and fungi in the air at each cultivation stage before the replacement of the HEPA filter was 169.7 CFU/m3 and 570 CFU/m3, and 126.3 CFU/m3 and 560 CFU/m3, during the medium production and harvesting processes, respectively. After the replacement of the HEPA filter, the bacterial density was the lowest in the incubation room and the fungal density was the lowest in the cooling room. The microbial populations isolated at each period consisted of seven genera and seven species before the replacement, including Cladosporium sp., six genera and six species after 1 month of replacement, including Penicillium sp., 5 genera and 7 species after 3 months of replacement, including Mucor plumbeus, and 5 genera and 12 species, 5 genera and 10 species, and 5 genera and 10 species, 4, 5, and 6 months after the replacement, respectively, including Penicillium brevicompactum. During the period after replacement, the species were diversified and their number increased. The density of airborne microorganisms decreased drastically after the replacement of the HEPA filter. Its lowest value was recorded after 2 months of replacement, and it increased gradually afterwards, reaching a level similar to or higher than that of the pre-replacement period. Therefore, it was concluded that replacing the HEPA filter every 6 months is effective for reducing contamination.

This study was carried out to investigate the changes in the biological activity and nutritional content of Flammulina velutipes, Grifola frondosa, and Sparassis crispa extracts after roasting treatment. Regarding biological activities, the DPPH radical scavenging activity was the highest in the extracts of air-dried G. frondosa, while the nitrite scavenging activity was significantly higher in the extracts of roasted S. crispa (p<0.05). The total polyphenol contents of F. velutipes and S. crispa were significantly increased by the roasting treatment compared with those in fresh samples (p<0.05). Regarding the amino acid composition of edible mushrooms, the content of sweet-taste amino acids, including serine (Ser) and alanine (Ala), increased in G. frondosa after roasting, whereas bitter amino acids, including decreased in roasted versus samples. Moreover, the contents of essential amino acids such as leucine (Leu), isoleucine (Ile), methionine (Met), valine (Val), and histidine (His) in F. velutipes and S. crispa were increased due to the roasting treatment (versus fresh samples). Thus, it was confirmed that the r method is effective in improving the nutritional content of edible mushrooms.
