Di-mono 교잡에 의한 잎새버섯 품종 "함박"의 특성 및 재배

A new commercial strain "Hambak" of Grifola frondosa was developed by di-mono crossing between dikaryon of ASI 9031 and monokaryotic strain derived from ASI 9021. It can be cultivated in plastic bottle and bag filled with oak and poplar sawdust substrate which supplemented with 15% rice bran or 10% corn bran. Improvement of cultivation methods was required for shortage of mycelium incubation period and management of humidity in growing room. The optimum temperature of mycelial growth was 25℃ and that of fruiting body development was 15∼18℃. The color of pileus surface was brown and fine pores, whitish to cream-colored, were underside. Primordia formation of Hambak was two days faster than that of Yipsae 1. The yield was 97g/ 850㎖ bottle, a bunch of fruiting body was bigger and the quality was better than Yipsae 1.

All intra-specific hybrids among ASI 2172 x 2104, 2172 x 2307, 2186 x 2172 and 2186 x 2307 in P. salmoneostramineus produced pink fruiting bodies as like wild parental types. However, three hybrids of them between ASI 2186 and 2104 were white fruiting bodies. A new commercial strain " Noeul " of Pink Oyster mushroom was developed by intra-specific hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from ASI 2172 and ASI 2104. The optimum temperature of mycelial growth and fruiting body development were 25∼30℃ and 19∼24℃, respectively. The pileus was bright reddish pink. Commercial strain "Noeul" was as prolific as the more commonly cultivated Pleurotus ostreatus in the conversion of substrate mass to mushrooms using bottle cultivation. Mushroom cultivator can save money for mushroom growing on summer in Korea. Mushrooms should be picked when moderately young, and handled carefully so as to not bruise the brilliantly colored gills. This pink color makes marketing an interesting challenge depending upon the market niche.

Analysis of genetic characteristics of Flammulina velutipes showed that strains have a range of 85% in genetic distribution diagram. According to this result, we divided these strains into five groups. In experiment of determinating the optimum media and condition in cultivating F. velutipes, we found the optimum temperature and pH range for hypha growth were 25℃ and 6.0 to 7.0, respectively. in addition, the best media for growth of that in plate was MCM (Mushroom Complete Media) which have a growth length from 68 to 83 mm. In vivo test, we observed that fast growth and good density of hyphae in mixture media of douglas fir sawdust, cotton seed meal and beet pulp (6:2:2 V/V). Also when we cultivated F. velutipes in this media, we harvested high yield of fruiting body.

느타리 신품종 "청산"의 재배 특성

A new variety of oyster mushroom 'Choengsan' was developed with crossing between monokaryotic strains derived from Suhan and Wonhyoung #3. It was developed by Agriproduct Processing Experiment Station, Gangwon Agricultural Research & Extension Services in 2006. The optimum temperature of mycelial growth was around 27∼28℃ and that of fruiting body was 14∼17℃. The color of pileus was dark-gray color. The period of primordial occurrence was about 20 days in bed cultivation using cotton waste and the yields were 38 kg/3.3㎡.

Penicillium rubrum에 의한 상황버섯(Phellinus baumii) 푸른곰팡이병의 발생

Green mold of Phellinus baumii mushroom caused by a species of Penicillium was observed in Daegu on August, 2000. The causal fungus was identified as Penicillium rubrum based on its cultural and morphological characteristics. Conidiophores of the fungus were one-stage branched and terminating in a whorl of 3∼5 metulae. The metulae were mostly 5.2∼7.8 ㎛ in size. Phialides were flask-shaped and 5.7∼7.5×2.2∼2.7 ㎛. Conidia were subglobose and 1.8∼2.3 ㎛. Colonies on Czapek's agar growing, attaining a diameter of 23 ㎜ within 8 days at 25℃. The optimum temperature for growth of the fungus was about 20∼30℃. This is the first report on the occurrence of green mold of P. baumii caused by P. rubrum in Korea.

This study for establishment of cultivation technique was carried out in home, Pleurotus ostreatus, Ganoderma lucidum. The mushroom kit allows you to grow some of the mushrooms used by the finest chefs in the world easily and quickly almost anywhere in your own home or office. Mushroom kits may be placed on a coffee table, counter, or desk. They will produce mushrooms virtually anywhere room temperature is maintained. Now we have the opportunity not only to enjoy watching these exotic mushrooms grow but also to enjoy eating the freshest organic mushrooms possible. To study for the possibility of indoor cultivation of P. ostreatus, and G. lucidum, we invested cultivation status with two kinds of sawdust, cotton waste and rice straw in washroom, kitchen, living room, and bedroom. It took 2∼4 days to make primordia formation in a block of P. ostreatus, and G. lucidum was 11 days. Weight of P. ostreatus on cotton waste, rice straw, and willow sawdust were 2,060g, 90g, and 770g, respectively. and weight of G. lucidum on oak sawdust was 172g. Th best result was achieved in washroom, among used washroom, kitchen, living room, and bedroom.

원형느타리버섯 백색돌연변체의 특성

The white-colored and the dark gray-colored mutants were frequently happened in cultivated areas of Pleurotus ostreatus (Wonhyeong-neutari). These caused conflicts between farmers and spawn companies. Our studies were conducted to elucidate the mechanism of mutagenesis. The results from the studies would provide valuable informations that could be used to prevent the color-related mutation, and also will be applied in breeding programs of P. ostreatus. Oyster mushroom variety, Wonhyeong-neutari, is somatic hybrid of Pleurotus and has genetic makers for arginine, ornithine, proline, riboflavine. Genetic markers analysis of monospore isolates derived from color mutants show identical tendency with that of Wonhyeong-neutari, these results indicate that color mutants were derived from Wonhyeongneutari. Twenty-one and four homokaryons were selected from the white-colored mutant MGL 2205 and gray-colored ASI 2029. All 34 F1 hybrids derived from the white-colored mutant MGL 2205 produce white-color fruiting bodies, indicating that the white color trait is heritable. In the first generation hybrids between the white-colored MGL 2205 and the gray-colored ASI 2029, all 16 hybrids produced pigmented fruiting bodies. Homokaryons isolated from the hybrid MGL 2205 X ASI 2029 were mated with homokaryon tester strains derived from MGL 2205. By these result, we could assumed that white color trait is a heritable character which is controlled by more than one recessive gene.

This study was carried out to characterize virus-like particles (VLPs) in Pleurotus ostreatus. The white and the dark gray mutants frequently observed in mushroom farms of Pleurotus ostreatus (Wonhyeong-neutari). A 5.8kb segments of dsRNA was detected only in the white mutants but not in the dark gray mutants. The VLPs were purified from the fruit bodies by Polyethylene Glycol (PEG) and ultracentrifugation. Electron microscopy analysis showed that VLPs were isometric about 14, 20∼45nm in diameter. Further study is needed to reveal the morphological and yield variations of mushroom strains including VLPs observed in the mushroom farms. Also it is needed to maintain fundamental research for taxonomy, diagnosis, and physiology of VLPs in the mushroom strains.

Application of sludge wastes into the field may help soil fertility with physical, chemical and biological properties. Efficient use of sludge waste, however, requires an individual assessment of the waste products. A lot of experiment into the organic waste-treated soils has been done for decade. However, studies have not been carried out on the assessment of agricultural soil by Basidiomycota. This study was assessed the influence of sludge application on soil healthy in agricultural upland soils. The organic wastes selected for long-term application experiment in this study were municipal sewage sludge (MSS), industrial sewage sludge (ISS), leather processing sludge (LS), alcohol fermentation processing sludge (FS), and pig manure compost (PMC). To develop the soil healthy assesment method, soil samples were diluted by 20X with distilled water. After shaking at 200rpm for 30 minutes, the shaked sample was mixed on PDA(Potato Dextrose Agar). And sterilized at 121℃ for 20 minutes. Coriolus hirsutus (MKACC 50560) was inoculated on petri-dish including PDA mixed sample. After the media was incubated at 25℃ for five days, the mycelial growth of C. hirsutus was measured. When the mycelial growth on sample media was compared with growth on media contained PDA only, well grown media contained sample soil was assesed as healthy soil. The results suggest that the simple method by Coriolus hirsutus is a handy way to assess the healthy of waste sludge-applied upland soils.
