Phellinus baumii 자실체 열수 추출물의 Sarcoma-180 에 대한 항암 및 면역효과

A hot water extract(HWE-P4) was separated from the fruit bodies of PMO-P4, and its antitumor and immunomodulatory activities against sarcoma-180 in ICR mice were investigated. The internal transcribed spacer (ITS) regions from PMO-P4 were amplified using polymerase chain reaction(PCR) and sequenced. The results revealed that PMO-P4 was belong to the Phellinus baumii. When oral administration at the dose of 160mg/kg/day in the mice until the end of the experiment with 2 week's pre-feeding of the HWE-P4, the survival rate of the mice was 152% for 50days after the inoculation of sarcoma-180 and the suppression rate of the tumor growth was 35.3%(p<0.05) for 28 days after inoculation of sarcoma-180. The HWE-P4 increased 71.4% of the CD4/CD8 ratio and 5-fold of the expression of CD25(IL- 2 receptor chain) compared with the control. From these results, the antitumor activity of HWE-P4 is exerted through its immunomodulating activity on the host's immune system.

We carried out survey of a pine mushroom picking district and its vegetation environment for increase and maintainment of its production in Kyonggi province. The district which we could pick up pine mushroom among the nineteen surveyed district were sixteen such as Geoleun ri Gangcheon myeon Yeoju gun Kyonggi do, etc. and total yields were 470∼600 kg in Kyonggi province. The temperature of atmosphere and earth in Yeoju and Bonghwa were droped to 19 at 5∼7th september and maintained from 12 ∼ 18 during the a pine mushroom picking period. The density of a pine tree per 100m2 was 5.1∼27.1 trees, the years of it was 15∼50years old and height was 4∼11 meter. The pine tree in a pine mushroom picking district was competed with oak trees, azaleas, lacquers, etc. The depth of organic materials accumulated on the pine mushroom mycelium layer was 4.5 cm in Yeoju. The soil texture of a pine mushroom picking district was almost sandy, soil pH was 4.3∼5.1 and organic material content was 3.91∼8.28%.

Mutual growth limitation was observation when the two antagonistic fungi was come in contact with each other. Brown line was formed 2day after contact with Trichoderma spp., and then, green spores formed overnight. The laccase activity of L. edodes was stimulated when this fungus wsa co-incubated with Trichoderma spp. for a few days in liquid media. In sawdust-rice bran nixtures, outstanding broun line developed when the two antagonistic fungi cocultured. The pH of the substrates changed from 5.5 to 4.5 after overgrowth, suggesting a difference in the degradation ability and the preference of the two fungi for the lignocellulose material.

The mycelial mass of K. mutabilis greatly increased at pH 5.5∼6.0 but decreased pH 6.0. The linear mycelial growth wsa mostly supported on sawdust of Quercus accutisima and the mycelial density wsa high on sawdust of Q. accutisima and corn cob. Much mycelial distribution could be showen in ray parenchyma cell and ray tracheid. Severe degradation of ray parenchyma cell was found but little degradation of ray tracheid cell was found. The dry weight loss wsa 5.9% after agar-block test. And the pH wsa acidified from 6.07 to 4.31 and hot water extractives was decreased after degradation of Q. serrata sawdust by K. mutabilis.

The optimal temperature for mycelial growth the F. fomentarius was 30 and the range of the temperature for mycelial growth wsa about 10∼30 . The optimal pH for the growth was 4.0. The percentage of weight loss percentage wsa 17.4%. The percentage of WEC extractives wsa increased to 2.24%. The observation of micromorphological showed that the detected cell wall were erosive and thinning as typical degradation pattern of whiterot fungi.

The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Fomitopsis pinicola are as follows. 1) Mycelial growth and density of F. pinicola. were the highest in the medium of PIDA(Pine Dextrose Agar; 66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA(pH 4.7), CHA, and MEA(pH 4.7). 2) Optimal temperature for the mycelial growth and density of F. pinicola were shown to be 30 , but the mycelia were dead at 40℃ . the mycelial growth and density of KNAC9005 strains was the highest at 30 (66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and 5℃. 3) Optimal pH for the mycelial growth and density of 40℃ was revealed to be 6.0(88.4mm/10d). above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of 40℃ were maltose(331mg/25ml/15d), peptone(347mg/25ml/15d), and glutamic acid(357mg/25ml/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40.

소나무잔나비버섯(Fomitopsis pinicola) 톱밥재배 연구

This study was carried out to investigate the physiological charateristics of F. pinicola in sawdust media. The optimum temperature in sawdust media was 30℃ in of F. pinicola. The optimum pH was 5 in F. pinicola. Mycelial growth and density of F. pinicola was quite good when birch tree and oak sawdust, respectively were used as cultural substrates. The best mycelial growth in F. pinicola was observed when beer waste was added as supplement on sawdust substrates. The optimum supplement ratios of beer waste and a magnecium sulfate were 20%, and 0.1% respectively. However, optimun supplement ratios of a calcium oxide and a LVD were different as 0.1% in F. pinicola.

Jinmi oyster mushroom(Pleurotus ostreatus) were bred in Mushroom Research Institute, Gyonggi Province A.R.E.S in 2003. This oyster mushroom were bred and cultivated one after mating single spores collected from K3-2 and ASI2018-249. The major characteristics of the mushroom are showing a lot of pinheadings, the gray- colored and infundibuliform pileus. The optimum temperature for the mycelial growth was around 25∼30℃ and that for the pinheading and growth of fruitbody was around 15±1℃. Around 22 days at 20℃ were required for incubation of Jinmi and the yield was shown high by 145.9g/bottle.

Sodam oyster mushroom(Pleurotus ostreatus were bred in Mushroom Research Institute, Gyonggi Province A.R.E.S in 2002. Sodam oyster mushroom were bred and cultivated one after mating single spores collected from Aenutari 1-ho and Suhan 1-ho. The major characteristics of the mushroom are showing a lot of pinheadings, the graycolored and shallow cornhead-shaped pileus. Around 20 days at 20 were required for incubation of Sodam and the yield was shown high by 148.2g/bottle. As the activity of hypha is so strong, the proper time of pinheading is demanded.

Hukbaek oyster mushroom were bred and cultivated one after mating spores from Aenutari 1-ho irradiated to X-ray and Byungnutari 1-ho. The major charateristics of the mushroom are showing that pileus is blackbrown and stems are pure-white and straight. The optimum temperature for the mycelial growth was around 25∼30℃ and that for the pinheading and growth of fruitbody was around 14∼16℃. Around 20 days at 20℃ were required for incubation time and the yield was shown high by 142.8g/bottle. When the cultivating temperature is over 17℃ the color of pileus becomes gray-brown.
