Antioxidant and inflammation inhibitory effects from fruiting body extracts of Ganoderma applanatum

In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.

Bacillus velezensis HKB-1 was isolated from the composted spent mushroom substrate of Lentinula edodes (LeCSMS) and inhibited mycelial growth of phytopathogenic fungal species, Phythhopthora capsici, Collectotrichum coccodes and Fusarium oxysporium by more than 70%. B. velezensis HKB-1 showed bacterial growth rate 10 to 100 times higher than that of other commercial bacterial media in water extract of LeCSMS supplemented with 1% molasses. The LeCSMS medium was effective in promoting the growth of pepper and controlling Phytophthora blight disease of pepper.

흰색 팽이 신품종 ‘설한’ 육성

Flammulina velutipes is the representative edible mushroom in Korea. It makes up around 18% of the total mushroom production and accounts for about 36% of the total mushroom export. Despite being one of the most industrially important mushroom in Korea, the distribution rate of domestic cultivar was low (~31.6%) in 2020. Therefore, we need a new domestic cultivar with high quality as the main export mushroom. We developed a new white domestic cultivar, ‘Seolhan’, bred by mating two monokaryotic strains isolated from Baekseung and Woori-1ho. The optimum mycelial growth temperature was 25oC and Seolhan had higher mycelial growth than the control at 20 and 25oC. In bottle cultivation, the optimum temperature of incubation was from 15oC to 17oC and the growth temperature of Seolhan was 7-8oC, which was slightly higher than that of the control. The yield of Seolan was 226.5±8.0 g, an increase of 18.4% compared to the control. The period of primodia formation also was a couple of days ahead. In morphological traits, the pileus of Seolhan was smaller than that of the control, the stipe was thicker, and the color brighter. As the results of replacement culture and genetic diversity test, it was confirmed that Seolhan was genetically different from other white varieties.

The purpose of this study was to identify and characterize the laccase genes of Flammulina velutipes var. lupinicola. Five laccase genes (g1934, g1937, g2415, g2539, g5858) were selected based on the copper binding site and signal peptide analysis results using the laccase gene selected from the F. velutipes var. lupinicola genome. The size of the laccase genes of F. velutipes var. lupinicola were 1,488 bp~1,662 bp. As a result of cDNA sequence analysis, 14 to 17 introns were identified in the laccase genes. The cleavage site predicted as the signal peptide of the laccase gene was found to be located between 20 bp and 34 bp from the N-terminus. In addition, separation and purification were performed to characterize the F. velutipes var. lupinicola laccases, and the optimal activity of the separated and purified proteins were analyzed by pH, temperature and time. Five bands with laccase activity were found from zymogram analysis. The optimal pH of the reaction was 5.5, the optimal temperature was found to be 40oC. Therefore, characterization of the laccase genes identified in this study should help in better understanding the biomass decomposition of F. velutipes var. lupinicola.

국내 유통 복령(Poria cocos)의 농약, 중금속, 방사선물질 및 회분 함량 조사 연구

The purpose of this study is to provide basic data on the safety of Poria cocos, a major Korean herbal medicine, and to establish tolerance levels for hazardous substances. Residual pesticides (321 types), heavy metals (7 types), radioactivity (3 types), and ash content were analyzed in 10 Poria cocos products distributed in Korea (5 domestic and 5 in China). Of the residual pesticides, cypermethrin was detected at 0.03 mg/kg in one domestic sample and was below the standard value. Of the heavy metals, the levels of Zn, Pb, Ni, Cu, Hg, and Cr were below the standard values in all samples. Hg was not detected in any sample. Cd was detected at 0.011 mg/kg in one Chinese sample. The radioactivity concentration was also below the MDA value in all P. cocos samples. The ash content was less than the standard value of 1.0% in all P. cocos samples. The findings indicate that P. cocos distributed in Korea is safe. However, because P. cocos is always exposed to harmful substances because of the nature of its cultivation in the soil, it seems necessary to establish systematic management standards for the entire production stage, such as cultivation, storage, and distribution.

This study was conducted to develop food and medicinal products containing useful components of Lentinula edodes in Codonopsis lanceolata and Platycodon grandiflorus for use as herbal medicine. We manufactured C. lanceolata (FCLM) and P. grandiflorus (FPLM) extract fermented with L. edodes mycelium. The effect of the two fermented products on proximate composition, free sugar, organic acid, β-glucan, ergothioneine, ergosterol, and vitamin D2 levels, and 3T3-L1 preadipocyte cell growth were studied. The proximate composition analysis results showed that the crude fiber and crude fat content in FCLM was higher than that in FPLM, and the crude protein and soluble nitrogen content in FPLM was higher than that in FCLM. Free sugar analysis detected arabinose, glucose, and sucrose in both FCLM and FPLM, and the total free sugar content was high in FPLM. The organic acid content was lower in FCLM and FPLM compared to C. lanceolata and P. grandiflorus before fermentation. The β-glucan content was higher than that of L. edodes used as a control in both fermented products, FCLM and FPLM. The content of ergothioneine, an antioxidant, was higher in FCLM than in FPLM. Ergosterol content was highest in L. edodes which was used as a control, and the two fermented products showed similar content. Vitamin D2 was detected only in FCLM and FPLM, and FPLM (0.58±0.01 mg%) showed a higher vitamin D2 content than FCLM (0.47±0.01). FCLM and FPLM showed a higher level of cell viability for 3T3-L1 pre-adipocytes compared to non-fermented C. lanceolata and P. grandiflorus. In addition, FCLM and FPLM inhibited 3T3-L1 preadipocyte differentiation more than C. lanceolata and P. grandiflorus before fermentation, which may exert an anti-obesity effect.

We investigated the effect of sodium chloride-associated abiotic stress on the development of Pleurotus ostreatus. We examined the growth characteristics of fruiting bodies, constituent amino acids, and fragrance pattern to determine the effect of culturing Pleurotus ostreatus on a sawdust substrate supplemented with sodium chloride in a dose-dependent manner. Pleurotus ostreatus fruiting bodies exhibited an increasing tendency towards augmented yields when grown in the presence of 0.5% sodium chloride as compared with that grown in the control group. However, increasing the supplementation of sodium chloride from 1.0 % to 2.0% resulted in significantly decreased yields of Pleurotus ostreatus fruiting bodies in these groups as compared with that in control groups. Further assessment revealed the presence of 14 types of amino acids in the fruiting bodies, including aspartate, threonine, serine, glycine, alanine, methionine, valine, isoleucine, leucine, phenylalanine, tyrosine, lysine, histidine, and arginine, at lower levels in all the sodium chloride-treated groups than in the control group; except for glutamic acid and proline. Similarly, fragrance pattern analysis of the Pleurotus ostreatus fruiting body by chromatography confirmed that the intensity of the substances presumed to be octane compounds, to which the unique flavor of mushrooms is attributed, was lower in all the sodium chloride-treated groups than in the control group.

Ligninolytic enzymes were produced by Pleurotus ostreatus No.42, cultivated in a new kind of bioreactor that has a rotating draft tube with a helical ribbon. Maximum laccase (Lac) production (about 8,200 U/bioreactor) was reached after 3 days of incubation, then production decreased. Production of manganese peroxidase (MnP) in this fermenter reached a maximum level of about 8,400 U/bioreactor after 6 days of incubation. Lignin peroxidase (LiP) was not detected under these growth conditions. These results indicate that the rotary draft tube bioreactor (RTB) is compatible with large scale production of ligninolytic enzymes. MnP produced under these fermentation conditions was purified via a multistep process that included chromatography on Sepharose CL-6B, prep grade Superdex 75, and Mono-Q. This major isoenzyme was confirmed to have an apparent molecular weight of 36,400 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and its isoelectric point (IEF) was determined to be 3.95. N-terminal sequencing of the major isoenzyme from this fermentation was identical to that reported for an MnP3 isoenzyme isolated under different cultivation conditions, including stationary and shaking culture.

갈색 느티만가닥버섯 추출물의 부위별 항산화 활성

This study was carried out to evaluate potential of Hypsizygus marmoreus (brown cultivar) as a functional food and drug materials. H. marmoreus were divided into pileus and stipe and extracted in hot water and 80% ethanol. The total polyphenol content was highest in the hot water extracts (pileus 17.15±0.19 mg of GAE g, stipe 7.37±0.16 mg of GAE/g) and pileus compared to the ethanol extracts (pileus 10.23±0.14 mg of GAE/g, stipe 3.76.±0.19 mg of GAE/g) and stipe. Also, hot water extracts of pileus from H. marmoreus (brown cultivar) was more effective DPPH, ABTS, ORAC value, reducing power than ethanol extracts and stipe extracts. The pileus and stipe extracts were confirmed to be non-cytotoxic in the mouse macrophage cell line RAW 264.7 determined by WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) assay. Overall, extracts of H. marmoreus (brown cultivar) was higher antioxidant activity than other mushrooms, and no cytotoxicity. Therefore, H. marmoreus (brown cultivar) showed potential as a functional food and drug materials. The brown cultivar of H. marmoreus have higher antioxidant activity than white cultivar, H. marmoreus seem to have different antioxidant activity depending on the cultivar.

UV-B조사 시간에 따른 흑타리 자실체의 저장 특성

This experiment was conducted to investigate the changes in freshness parameters and determine the optimal UV-B irradiation conditions for each storage period for Heuktari, an oyster mushroom cultivar, grown under LED lightNo significant changes were observed in the CO2 and O2 content until day 12 of storage; however, after day 15, the CO2/O2 ratios were higher in the UV-treated groups than in the untreated groups. No color difference was observed between the UV-treated and untreated groups. In all UV-treated groups, hardness decreased over time; however, the group irradiated for 15 minutes showed higher hardness values t han those of the untreated group. The UV-irradiated group showed higher ergosterol content than did the untreated group, and scanning electron microscopy revealed that mushroom cell wall destruction increased with increased UV treatment time. Overall, these findings confirmed that UV-B irradiation increased the ergosterol content of Heuktari but decreased its freshness and negatively impacted other storage characteristics.

The cleaved amplified polymorphic sequence (CAPS) marker uses a restriction enzyme recognition site resulting from single nucleotide polymorphisms and insertions and deletions on the DNA sequence. This technique does not require expensive equipment, the process is simple, and clear results can be obtained reliably. In this study, Agaricus bisporus cultivars SaeA, SaeDo, SaeHan, SaeYeon, SaeJeong, Dodam, Seolgang, Dahyang, Hogam, and Hadam developed in Korea were discriminated using four CAPS markers. Our results indicated that it is possible to distinguish the ten cultivars and determine the genetic diversity among them.

산느타리(Pleurotus pulmonarius) 품종의 초위성체(simple sequence repeats) 특성구명

Simple sequence repeats (SSRs) were isolated from major Pleurotus pulmonarius cultivars in Korea, namely ‘HS47’ (monokaryon, gamete of ‘Santari’), ‘GB19’ (monokaryon, gamete of ‘Santari’), ‘Hosan,’ ‘Yeoleumneutali1,’ ‘Sambok,’ ‘Gangsan,’ ‘Yaksan,’ ‘Jasan,’ ‘Hyangsan,’ and ‘Yeoleumneutali2,’ and characterized via HiSeq genome sequencing and bioinformatic analysis. The genome sizes of the monokaryons ‘HS47’ and ‘GB19’ were estimated to be 37.3 and 37.2 Mb, respectively, and those of the other dikaryotic cultivars ranged from 47.1 to 61.1 Mb. A total of 711 (smallest) and 1,106 (1.5 times the smallest) SSRs were found in the ‘HS47’ and ‘Gangsan’ genomes, respectively. Hexanucleotide and octanucleotide motifs accounted for the top two fractions of all SSRs. CGA/TCG, A/T, and CTC/GAG were the most frequently detected nucleotides in the SSRs. Most of the SSRs were 21~30 nucleotides long (hypervariable for application), accounting for 70% of all lengths of SSRs.

국내 표고(Lentinula edodes) 주산지인 장흥지역에 적합한 국산 원목품종 선발

In this study, to select suitable shiitake strains in Jangheung region for domestic strains of Lentinula edodes for log cultivation, a total of five different Korean strains(Chunbaegko, Sanjo 303ho, Poongnyunko, Baekhwahyang, Soohyangko) and two Japanese strains (Mori 290, Moriyujiro) were selected and a cultivation test was conducted to select suitable strains. As a result of the yield, Sanjo 303ho produced 40.9 kg, which was superior to 39.2 kg for Mori 290 and 37.7 kg for Moriyujiro. As for the quality characteristics, it was confirmed that Baekhwahyang had a hemispherical shape, and high quality fruit bodies. In conclusion, Sanjo 303ho, which has excellent production, and Baekhwahyang, which is advantageous for producing high-quality shiitake, were selected in Jangheung region for domestic strains.
