This study were conducted to investigate pin-heading induction and growing temperature of Agaricus brunescens. The temperature for fruit body formation was better at 15℃ than at 18 and 21℃. As the temperature go up to 21℃, the fruit body number was decreased but the fruit body size and weight was increased. Optimal temperature for both pin-heading and growth of Agaricus brunescens was 18℃.

This study was carried out to investigate effect of NaCl treatment on growth oyster mushroom and Trichoderma spp. on PDA and in rice straws. We also investigated the possibility of oyster mushroom cultivation using rice straw harvested from the reclaimed land having been drained sea water. Mycelial growth of oyster mushroom was increased by treatment of 0.2% NaCl but decreased by treatment of the higher concentration of NaCl. In the case of the mycelial growth on PDA of Trichoderma spp., no change was found in 0.5%∼1.0% NaCl, but decreased in the range of 1.0%∼3.0% NaCl and drastically decreased at 5.0% NaCl. In the rice straws treated with different concentration of NaCl solution, mycelial growth of oyster mushroom showed almost same result compare to PDA. The spore formation of Trichoderma mould was almost same in both 0% and 0.3% NaCl, decreased in 0.5% and was not found spores in the higher concentration of NaCl. As increasing salt concentration in the rice straws, the NaO contents were increased. The K2O contents were decreased before and after sterilization. The moisture content of rice straws showed no difference by treatment of 3.0% NaCl but decreased the moisture by treatment of 5.0% NaCl. No pH change was found in the rice straws treated with NaCl. Mushroom yield in the rice straw of reclaimed land was a little higher than that of normal paddy land straw. The duration of primordium formation was not affected by NaCl concentration in rice straws. The yield of fruiting body in 0.3% NaCl treatment was 2,700kg, which was almost same to non-treated plot, but decreased in 0.5% NaCl. EC value of soaking water after submerging rice straw was higher than before, but NaO content was not changed at both condition.

Effect of solid grain media on the mycelial growth of Pleurotus ostreatus and its biofunctional activities were investigated using seven kinds of grains. Foxtail millet and barley were good for growth of hypha of P. ostreatus. However, growth of the mycelium was very slow in the solid grain media containing wheat, corn and brown rice. Mycelial growth of P. ostreatus according to water content of solid grain media was good at 25% to 30%. Mycelial growth of P. ostreatus according to heating-time and temperature of solid grain media was good for 30 min at 121℃. Anticancer activities against lung cancer cell line of the myceial extracts from P. ostreatus grown on several grain media were strong in the corn, defatted soybean, brown rice, barly and black bean in order. Fibrinolytic activities of the myelial extracts were strong in order defatted soybean, wheat, foxtail millet, barley, brown rice and black bean. The mycelial extracts were showed good antibacterial activities against Pseudomonas aeroginosa and Staphylococcus aureus.

국내에서 많이 재배되는 버섯 중의 하나인 느타리 P. ostreatus는 먹물버섯이나 치마버섯과는 달리 실험실 수 준에서의 자실체 발생이 힘든 것으로 알려져 있다. 본 연구 에서는 실험실에서 느타리버섯을 샤레를 이용하여 전 발 생과정을 유도하기 위한 방법을 검토하였다. 샤레상의 배 양조건은 프라스틱 샤레 (60×15 mm)의 감자배지에서 균사를 접종한 후 빛이 없는 상태에서 배양한 뒤에, 균사의 환기상태, 균사표면의 상처, 빛 그리고 저온충격 등의 여러 환경요인들이 원기 및 자실체 형성에 미치는 영향을 검토 하였다. 느타리의 최초 자실체 형성은 접종 이후 3주 내에 얻을 수도 있었으며, 균사접종 이후 10주 동안에 자실체의 형성은 균주에 따라서 76%에서 97%의 높은 빈도로 유도 될 수 있었다. 위와 같이 샤레상에서 자실체를 형성 할 수 있었으며, 정상적인 자실체의 성장을 위해서 빛은 필수적 이며, 환기도 필요하였다. 또한 균사의 상처 처리가 원기, 자실체 및 포자의 형성에 미치는 영향이 균주에 따라서 크 게 차이가 났으며, 같은 균주라 하더라도 발생단계별 그 반 응의 차이도 크다는 사실을 확인하였다. 이들 자실체에서 수집된 담자포자는 발아가 가능하였다. 샤레상의 완전한 자실체 형성 방법은 느타리버섯의 생육주기를 단축시킬 수 있을 뿐만 아니라, 다양한 재배조건의 시료 획득과 한 개의 배양 용기 내에서 버섯 발생의 전 단계를 관찰하고 분 석하는데 유용할 수 있다.
Pleurotus ostreatus, the oyster mushroom, is one of the most widely cultivated and important edible mushrooms in the world. In order to study the developmental process of P. ostreatus and its regulatory mechanism, a new culturing method needs to be established for inducing the fruiting body and sporulation in the laboratory. In this study, we have examined whether the fruiting body of P. ostreatus can be formed on the plastic petri dish which are commonly used for cell culture in the laboratory. The strain was cultured on 60×15 mm plastic petri dish with potato dextrose agar media at 28℃ for mycelial growth and then at 18℃ for the formation of primordia and fruiting bodies within plant growth chamber. The development of primordia into fruiting bodies was achieved on cultured dishes under air ventilation. At the primordia stage, the normal formation of fruiting body was blocked by sealing the plastic dish with parafilm. The periods requiring for the formation of primordia and fruiting bodies were examined on the dish culture. About 96% and 76% of cultured samples formed primordia and fruiting bodies under the optimal conditions during ten weeks of culture, respectively. These culturing periods, however, were changed by the mechanical injury treatment to mycelia. As other factors affecting the fruiting body formation, the effects of light and cold shock have been tested. No fruiting formation was observed on the cultured dishes under the dark. The cold shock treatment by storing cultured dishes for one day at 4℃ did not have any significant effects in the fruiting body formation. Spores of fruiting bodies acquired from the petri dishes could be germinated on culture media at 28℃. These results suggest that the fruiting bodies of P. ostreatus can be formed on the experimental petri dish and this dish-culturing method is useful for understanding of the developmental process of P. ostreatus in the laboratory. Furthermore, the dish-culturing method is able to shorten the life cycle of P. ostreatus without requiring large area and expensive device.
눈꽃동충하초(Paecilomyces tenuipes)의 균사생장조건 구명

Distribution and in vitro growth characteristics of entomopathogenic Paecilomyces species were studied based on the specimens and isolates deposited in Entomopathogenic Fungal Culture Collection (EFCC), Kangwon National University, Chuncheon, Korea. Paecilomyces species were frequently collected from different mountains and valleys and were almost distributed uniformly. Collection period of Paecilomyces species ranged from July to September, with most of the collections at the end of September. P. tenuipes was mostly collected one among different Paecilomyces species. The optimum medium for mycelial growth of P. tenuipes was PDA, whereas in other media such as SDAY and HMA also produced similar growth. The optimum temperature and pH levels were 25C and 6.0~7.0, respectively. Best carbon and nitrogen sources were dextrose and tryptone, respectively, while KH2PO4 was the best mineral source for mycelial growth. Highest mycelial growth was observed when the C/N ratio was 20:1.

This experiments was conducted to study on the substitute cotton wastes for ricestraw compost media on cultivation of Agaricus bisporus. This results were as follows. The results of yield according to the mixture rate of cotton wastes(4000~5500kg/198㎡), rice hull(4.5%), chicken manure(50%), gypsume(1.5%), urea(2%), tobacco powder((2.5%), seasame meal(5%) and MgSo4(5%) was effected to 57.2% increasement with 64.33kg in summer season, 65.21kg in winter season, 39.6kg in control, respectively. The results of yield according to the mixture rate of cotton wastes(4000~5500kg/198 ㎡), cotton hull(1.25%), rice hull(4.5%), chicken manure(50%), gypsume(1.5%), urea(2%), cigarette powder(2.5%), seasame dregs(5%) and MgSo4(5%) was effected to 62.3% increasement with 61.0kg in summer season, 62.3kg in winter season, 39.6kg in control, respectively. The results of yield according to the mixture rate of cotton wastes(2000~2500kg/198㎡), cotton hull((2000~2500kg / 198㎡), rice hull(2.5%), chicken manure(37.5%), gypsume(1.5%), urine(2%), cigarette powder((1.25%), seasame dregs(2.5%) and MgSo4(3%) was effected to 40.4% increasement with 57.33kg in summer season, 58.34kg in winter season, 39.6kg in control, respectively.

To develop neohaplonts for Ganoderma breeding, protoplasts were isolated from dikaryotic mycelium and regenerated. Selection rate of neohaplonts varied between ASI7074, ASI7091, ASI7094, ASI7100 and ASI7115, showing 5.24% on the average. Auxotropic mutants from Ganoderma monokarions were recovered by UV irradiation on protoplasts. Protoplast survival rates were 1.9% ASI 7074, 0.17% ASI 7091, and zero percent ASI 7100 using 300 second irradiation. Four auxotrophic strains were recovered from 1,536 colonies screened that will be further utilized for protoplast fusion and transformation.

Compatibility test of the 115 world-wide collections of Ganoderma strains including 61 of Korean origin, were performed by di-mon mating with tester strains G. lucidum G001-1 (A1B1) and G. tsugae 1109-16 (A1B1) . Unexpected results were found in that 75 strains, formerly considered to be G. lucidum exhibited clamp connections in mating with the tester 1109-16. This observation suggests that these strains require further examination prior to taxonomic classification. Interestingly, this compatibility could be confirmed more easily by the finding of distinct confrontation line between two strains on plate.
Reduction of labor in oyster mushroom cultivation by vinyl mulching on mushroom bed

Labor required for managing and harvesting the oyster mushroom bed was evaluated. Although vinyl mulching cultivation method needs more hours for spawning, it saves more than 50% of labor for harvesting and managing of the mushroom bed. Harvesting hour of 1st-3rd flush in vinyl mulching method was 48~50% for Plerutus ostreatus and 36~41% for P. sajor-caju. Labor for bed management after harvesting in vinyl mulching method was also 38~50% for P. ostreatus compared to conventional method, and 20~35% for P. sajor-caju. Vinyl mulching is believed to be a very efficient method for saving labor in oyster mushroom cultivation.
