1)Glycosylation is a key mechanism in determining diversity of natural products, and influencing their bioactivities. This approach requires a core set of glycosyltransferase that synthesizes the diverse sugar structures observed in nature. Recently, the researchers have begun to alter the sugar moiety and glycosylation patterns of natural products both in vivo E. coli system and in vitro for their glycodiversification. This review highlights new glycosylation tools using microbialproduced deoxysugar and a flexible glycosyltransferase on natural plant-flavonoids to generate novel glycoforms with useful biological activity.

항균물질을 생산하는 토착 미생물 Paenibacillus sp. BCNU 5011의 특성화

Strain BCNU 5011 was isolated from forest soil samples collected in the Taebaek mountain in the Gangwon province, Korea. The biochemical, physiological and 16S rRNA sequence analysis strongly indicated that this isolate was most closely related to Paenibacillus polymyxa. A maximum production level of antimicrobial substances of Paenibacillus sp. BCNU 5011 was achieved under aerobic incubation at 30℃for 3 days in SST broth.Paenibacillus sp. BCNU 5011 showed a broad spectrum of activity against Gram positive and Gram negative bacteria, including methicllinresistant Staphylococcus aureus (MRSA). Paenibacillus sp. BCNU 5011 was also shown to inhibit the growth of different potential human pathogenic bacteria and fungi in vitro. Peptide extract showed better antimicrobial activity than solvent extracts. But active antimicrobial compounds might be included in both peptide extract and solvent extracts. Further separation, purification and identification of active principles leads project to develop antimicrobial agents and anti-MRSA agents

Sodium butyrate에 의한 돼지 전염성 위장염 바이러스 백신의 생산성 향상

1)The essential operating parameters in virus vaccine production are multiplicity of infection (MOI), harvest time, and infection time. Stimulating agents also can be applied in order to improve vaccine productivity further. We investigated the optimum operating conditions in porcine transmissible gastroenteritis virus (TGEV) vaccine production and the applicability of sodium butyrate (NaBu) as a stimulating agents for the improvement of vaccine productivity. The optimum MOI, infection time, and harvest time for high production of TGEV by swine testicle (ST) cells were found to be 0.0001 pfu/cell, 3 day after cell inoculation, and 24 hpi, respectively. NaBu is known as a histone deacetylase inhibitor that has been widely used for the high expression of recombinant protein using mammalian cells and for the enhancement of virus propagation. So we tried to examine the potential of NaBu as a stimulating agent and to determine the optimum concentration by comparing TGEV titers with different range of NaBu concentration. TGEV titer with 5 mM NaBu was 1.5 times higher than control. Therefore, we concluded that NaBu can be a promising agent for stimulating various vaccine production including TGEV and the optimum NaBu concentration for TGEV production was determined to be 5 mM.

다시마 열수추출물의 성분 및 항산화활성 측정

1)Contents of amino acids, vitamins, and minerals as well as its antioxidant activitiy of Laminaria japonica water extract have been analyzed for preparation of functional foods and cosmetic products. From the analysis of total amino acids, eighteen kinds of amino acids were found in the water extract of Laminaria japonica. Among total amino acids, the order of contents was glutamic acid (2.07 mg/g), alanine (0.51 mg/g), aspartic acid (0.44 mg/g), glycine (0.34 mg/g), and valine (0.34 mg/g). In case of free amino acids, glutamic acid (0.95 mg/g), prolin (0.54 mg/g), aspartic acid (0.44 mg/g), leucine (0.07 mg/g), and phenylalanine (0.07 mg/g) were dominant compositions. Vitamin E was only detected in water extract of Laminaria japonica. The mineral contents were as follows: K 752.60 mg, Na 259.20 mg, Ca 80.20 mg, P 29.50 mg, and Fe 8.32 mg based on 100 g Laminaria japonica water extract. The nitrite scavenging activity of the extract were gradually increased with the extracts contents to 86.2% at concentration of 100 mg/mL and DPPH radical scavenging activity of the extracts were 86.4% at concentration of 50 mg/mL.

효모에서 생산한 재조합 human L-ferritin의 생화학적 특성 및 나노입자의 철산화물 합성

1)In the synthesis of nanoparticles, much attention has been paid to regulating the particle size. There has been a possible evident that using the central cavity (core) of the protein ferritin has a greatly significant influence on it because the core can generate the nanometer-sized mineral particles of variable metal ions. In this report, recombinant human L-ferritins produced from Saccharomyces cerevisiae were purified and their molecular properties were characterized. The cDNA for human ferritin L chain was also expressed in another host such as Escherichia coli, and the properties of recombinant L-ferritins were compared. From isoelectric focusing experiment, the L-ferritin from the recombinant yeast showed no indication of N-glycosylation. Some post-translational modifications other than N-glycosylation were speculated in the L-ferritins from yeast. A difference was made in the L-ferritins in their iron uptake rates and the initial rate of the L-ferritin from yeast was slightly increased. The reconstitution yield and size distribution of the core minerals were analyzed in the L-ferritins by transmission electron microscopy. The L-ferritin from yeast with higher reconstitution yield (54.5%) showed slightly larger sizes (mean 6.92 nm) with narrower size distribution than the L-ferritin from E. coli. It is, in conclusion, speculated that L-ferritin from yeast is relatively superior to the other, in view of the size of nanoparticle and its relative homogeneity.

1)To investigate the antihyperglycemic and antihyperlipemic effect of 80% ethanol extract of Schizandra Chinensis fruit, we induced diabetes in the rats with streptozotocin (STZ) and administered schizandra extract or Acarbose to diabetic rats for 21 days by oral administration. Consequently, the groups treated using schizandra extract decreased blood glucose levels more 39% than no treatment group and the case of Acarbose group was decreased it about 21%. The concentration of cholesterol, triglyceride and LDL-C in blood was also decreased while treating schizandra extract, on the other hand, HDL-C concentration was significantly increased it about 26%. Those results induced that anti-atherogenic index (AAI) in blood was improved more than 82% level like normal condition, especially in treatment of schizandra extract 100 mg. The lipid profiled in feces was likewise showed apparent tendency to decrease and food efficiency ratio of diabetic rats was became higher for treatment with schizandra extract, but Acarbose group had low efficiency in compared with the result of glucose level and lipid profile in blood. As a result, schizandra extract is regard a good medicine for diabetes due to improve physical constitution, blood glucose and lipid level caused hyperglycemia and suggest that schizandra extract has real effects on the diabetes complication as atherosclerosis, coronary heart disease, high blood pressure.

생물정보학적 접근을 통한 Caenorhabditis elegans 모델시스템의 생체내 RNAi 기능예측 및 비특이적 공동발현억제 현상 분석

Some computational approaches are needed for clarifying RNAi sequences, because it takes much time and endeavor that almost of RNAi sequences are verified by experimental data. Incorrectness of RNAi mechanism and other unaware factors in organism system are frequently faced with questions regarding potential use of RNAi as therapeutic applications. Our massive parallelized pair alignment scoring between dsRNA in Genebank and expressed sequence tags (ESTs) in Caenorhabditis elegans Genome Sequencing Projects revealed that this provides a useful tool for the prediction of RNAi induced cosuppression details for practical use. This pair alignment scoring method using high performance computing exhibited some possibility that numerous unwanted gene silencing and cosuppression exist even at high matching scores each other. The classifying the relative higher matching score of them based on GO (Gene Ontology) system could present mapping dsRNA of C. elegans and functional roles in an applied system. Our prediction also exhibited that more than 78% of the predicted co-suppressible genes are located in the ribosomal spot of C. elegans.

1)The chromatographic enantiomer separation of 9-anthraldimine derivatives of α-amino acid methyl and ethyl esters on four polysaccharide based chiral columns was performed. The 9-anthraldehyde Schiff base derivatives of α- amino acid esters were readily synthesized by stirring the α- amino acid ester hydrochloride salts with 9-anthraldehyde in the presence of 1,8-diazabicyclo [5.4.0]undec-7-ene as a base and anhydrous MgSO4. Chiralcel OD or Chiralcel OD-H showed the greatest enantiomer resolution of 9-anthraldimine derivatives of α-amino acid methyl and ethyl esters. The L-enantiomers of all the analytes were preferentially retained on Chiralcel OD or Chiralcel OD-H. This analytical method was applied in the determination of optical purities of several commercially available D- or L-α-amino acid methyl esters.

화학응집제를 이용한 미세조류의 응집 특성

1)The aim of the study was to optimize harvesting method for concentrating microalgae from microalgae mass culture. It is well known that the mass density of microalgae is usually very low and these are small size (5-20 μm) in the culture medium. It is essential that microalgae is harvested and concentrated economically for economical biodiesel production from microalgae. In this study, to determine optimized conditions for microalgae harvesting by chemical flocculation. Flocculation of three algae, Chlorella ellipsoidea, Dunaliella bardawil, and Dunaliella tertiolecta, was performed using various chemical flocculants, such as inorganic flocculants (aluminium sulfate, aluminium potassium sulfate, ferrous sulfate, ferric sulfate, ferric chloride, calcium hydroxide, sodium carbonate, sodium nitrite, and sodium aluminate), organic flocculant (polyacrylamide), and biopolymer flocculants (chitosan and starch). The results indicated that aluminium based inorganic flocculants is suitable for microalgae harvesting such as Chlorella ellipsoidea, Dunaliella bardawil, and Dunaliella tertiolecta. The results also recommended that flocculant doses, agitation speed, agitation time, sedimentation time for economical microalgae harvesting method using chemical flocculants.

미역 열수 추출물의 항산화 및 아질산 제거효과

1)For the development of functional food and cosmetics using the hot water extract of Undaria pinnatifida, the concentrations of vitamin, amino acid and element and activities of antioxidant and nitrite scavenging were investigated. The results are shown as follows: Vitamin C and E concentrations were 0.301 and 0.11 mg/100 g, respectively. Mineral concentrations were an order of Ca > Mg > K > Fe. The concentrations of total amino acids were an order of Glu > Ala > Val > Leu > Gly > Pro. Total phenol concentration and DPPH radical scavenging activity were increased with the increase of the concentration of extract. Especially, when the extract concentrationwas increased from 1.0 to 10.0 mg/mL, the total phenol concentration was increased from 0.043 to 0.125 OD 725 nm. DPPH radical scavenging activity at 50 mg/mL was 70.1%. The antioxidant activity of extract was stable in range of 80 to 140℃ and pH 3-9. The nitrite scavenging activity was increased with the decreaseof pH and the increase of it’s extract concentration. Especially, it was 83.4% at 50 mg/mL (pH 1.2). These results showed that the hot water extract of U. pinnatifida can be applied to functional food and cosmetics.

면역글로불린제제 효능평가를 위한 5종 B형간염 표면항원항체검출법의 비교

Five assays for anti-HBs were compared to improve potency test of Human lgG preparations. The three commercial EIA kits were optimized including dose response curve ranges and compared by conducting a co-laboratory study. After selecting the most reproducible EIA kit, methods comparison was performed with 22 samples in 5 different days. As a result, EIA (7.7 ± 5.3%) and MEIA (AxSYM: 3.7 ± 1.9%, IMx: 1.6 ± 0.8%) showed precision and accuracy (100.1 ± 12.6%). Therefore, the validated EIA assay was established and it is believed to be comparable to current MEIA.

사과꽃잎 추출물의 용매 분획으로부터 기능성 소재의 탐색

Fractional solvent extraction by organic solvents such as hexane, chloroform, ethylacetate, and butanol was carried out using 70% ethanol extract of apple flower leaves. Biological activities including antioxidant, whitening, antimicrobial and anti-wrinkle activities were investigated and bio-active materials of the extracts were identified using GC/MSD. Among the tested solvent fractions, ethylacetate fraction showed the highest total polyphenol content (1218.94 μg/mL), and flavonoid (140 μg/mL). The DPPH radical scavenging activities was over 80% at a dry matterbased concentration of 200 μg/μL and SOD-like activity was over 90% at 50 μg/mL concentration in ethylacetate fraction that was slightly lower than of ascorbic aicd. Tyrosinase inhibition activity related to skin-whitening was over 60% by ethylacetate fraction of 100 μg/mL. As an anti-aging effect, elastase inhibitory activity was about 45% in ethylacetate fraction. Also, it showed a significantly antimicrobial activity against P. acenes. From GC/MSD analysis, a characteristic peak of high content in ethylacetate fraction was identified as kaempferol, which has been reported as a bioactive compound.

iNOS 발현 검출을 위한 in vitro 시스템의 확립 및 적송잎 추출물에 의한 저해효과 검증

This study was aimed to verify suppressive effect of pine-needle extract on lipopolysaccharide (LPS)-induced inducible nitric oxide synthase (iNOS) expression. In order to evaluate suppressive effect on iNOS expression, RAW 264.7 cells were stably transfected using an iNOS promoterluciferase reporter plasmid yielding RAW 264.7/pGL2-NeomiNOS_ pro11 cells. Established in vitro detection system revealed to diminish LPS-induced iNOS expression by 0.1~500 μg/mL of saponin at the concentration-dependant manner. Pine needle extract also diminished LPS-induced iNOS expression to 92 and 88% at 500 and 50 μg/mL, respectively. These results suggest that the in vitro detection system developed here could be useful for the verification of suppressive materials on iNOS expression and pine needle extract could be used for the development of functional foods.

Headspace 법을 이용한 생굴 및 굴 자숙액의 감마선 조사에 의한 휘발성 냄새 성분 변화 분석

This study was conducted to investigate the effects of gamma irradiation on the change of volatile flavor compounds of raw oyster and its cooking drips using headspace methods. Major volatile flavor compounds of the raw oyster were identified as methylthiomethane and 1,5-hexadiene. When the raw oyster was irradiated at the dose of 5 kGy, 1-pentane was newly detected. On the other hand, 9 compounds including N-methoxyformaldehyde were identified as the major volatile compounds of cooking drips from oyster. Among them, N-methoxyformaldehyde contents in cooking drip was decreased by the gamma irradiation. By the gamma irradiation above 30 kGy, new heterocyclic compounds was found in oyster cooking drips. Therefore, the amount of volatile flavor compounds in the raw oyster and cooking drips were changed by gamma irradiation, and these results could be potentially used in the seasoning industry.
