Dictyophora indusiata is an edible mushroom belongs to Family Phallaceae of Phallales, Basidiomycota. The purpose of this study was to investigate the antioxidant and anti-inflammatory activities of methanol and hot water extracts prepared from fruiting bodies of Dictyophora indusiata. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity for those extracts. To measure the anti- inflammatory activities for the extracts, nitric oxide(NO) production from lipopolysaccharide(LPS) treated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that the extracts have excellent DPPH scavenging and chelating activity on the ferrous ions compared with positive control. The nitric oxide (NO) production in LPS-stimulated RAW 264.7 macrophage cells were decreased as we increased the concentration of the mushroom extracts. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/ kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. Therefore, the experimental results suggested that methanol and hot-water extracts of Dictyophora indusiata fruiting bodies might be used for natural sources of antioxidant and anti-inflammatory agents.

Characteristics of artficially cultivated Ganoderma applanatum fruitingbody KCI 등재

Ganoderma applanatum is one of the most popular medicinal mushrooms due to the various biologically active components it produces. In order to study for the possibility of artificially cultivating G. applanatum, we investigated the growth status of G. applanatum mycelium using oak sawdust bottle(850 ml). It took approximately 17~27 days for G. applanatum cultivation and 15~16 days for G. lucidum cultivation with sawdust until the mycelial growth was complete. The weight of dried fruiting bodies showed that G. applanatum GBGA-01 weighed 16.0±7.4 g, and G. lucidum ASI 7125 weighed 7.9±2.7g. The thickness of the pileus was measured as 10.6±2.6 mm for G. applanatum GBGA-01, 16.3±8.7 mm for G. applanatum ASI 52823 and 8.6±3.5 mm for G. lucidum ASI 7125. The color of Ganoderma spp. showed that L degree was 55.5±1.1 for the contex of G. applanatum GBGA-01, 47.7±2.2 for the contex of G. applanatum ASI 52823 and 39.6±2.9 for the contex of G. lucidum ASI 7125.

P. linteus (Korea Sanghwang, PLKS) and P. baumii (Jangsoo Sangwhang, PBJS) were artificially cultivated under the same conditions. Fruiting bodies were extracted using 70% methanol, 60% ethanol, and hot water. Phenol and flavonoid contents were optimally extracted using 60% ethanol. Antioxidant activities of 60% ethanol extracts from fruiting bodies and mycelia from each Sanghwang mushroom species were measured using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azino-bis (3- ethylbisthiazoline-6-sulfonic acid) radical scavenging activities, and FRAP (ferric reducing antioxidant power). The antioxidant activities of fruiting bodies were relatively higher in comparison to those of mycelial samples. In high performance liquid chromatography (HPLC) analysis, styrylpyrone-class poly phenolic compounds, davallialactone, hispidin, hypholomine B, and inoscavin A were detected in fruiting body samples of two Sanghwang mushroom species, but not in their mycelial samples.

The purpose of this study was to evaluate the mineral difference about Phellinus from cultivated on mulberry, oak and elm by ICP test. The major mineral components were Ca, K, Fe, P, Al and Na. In Ca, fruiting bodies from cultivated on mulberry were shown about 2~4 times highest, and in case of K was fruiting bodies from cultivated on oak shown about 2 times highest as against the other sample groups. Especially, Na was fruiting bodies from cultivated on oak shown about 10 times highest as against the other sample groups. In this study, fruiting bodies on P. linteus and P. baumii from cultivated the same kind of material lumber are similar mineral components, and we can’t really find much their difference . As a results, what kind of mineral component content had showed depend on material lumber.

Cellulose is the most abundant organic polymer constituent of the cell wall of green plants and of various forms of algae. The complexity of lignocellulosic biomass is a major challenge in industrial research. Most mushroom species that naturally grow on soil or wood possess cellulases and the corresponding enzymatic system and, potential candidates for the direct bioconversion of softwood polysaccharides into fermentable sugars. However, there have been fewer studies on mushroom cellulases than on fungi such as Trichoderma spp., exploit the full potential of mushroom cellulases. This review will focus on the current status ofmushroom cellulase research and applications and will provide insight into promising future prospects.

The cultivation of Poria cocos on tree-stumps and logs, was first described at the beginning of the 13th century. Poria cocos has been used for medicinal purposes to treat physical and mental recuperation, promote diuresis, forgetfulness and physical weakening. The artificial cultivation techniques of P. cocos was reported in China in 1957. are reported. The basic study on morphological characteristics and artificial cultivation method of Poria cocos with pine tree log buried under ground were initiated by Rural Development Administration. To widen the utilization of harvested sclerotia of Poria cocos as new food stuffs, powder of the sclerotia were used for producing new soft drinks and supplemented to wheat flour to make functional bread. ○ Bokryeong tree cultivation GB2, GB3, GB4, GB5, GB7, GB8, GB9, GB10, Bekryeong 1, Andong 1 and Andong 3. It is the result of investigation on the characteristics of the aged dates derived from the encapsulation cultivation of Bykryeong. As a result of measuring the number and weight of fruiting bodies, relatively large elongation was induced in Bokryeong No.1 and crossed strain GB4, and the L value of GB9 was measured relatively high in the skin color of fruiting body. The cut surface chromaticity was measured to be low in GB5. ○ Glass bottle production The result of inducing the aging in the glass bottle was as follows is the photograph of the pine tree in the glass bottle, and taken after removing the glass bottle. Crosses GB1-10 and Bokryeong No.1, Korea Forest Service 1104, Korea Forest Service 1105. The weight, size and chromaticity of the pine trees were investigated. As a result of measuring the number and weight of fruiting bodies, GB3 and GB4 produced comparatively large agglomerates and the fruity body skin color GB7 was high.

Bioactive polysaccharides isolated from the fruiting bodies of Phellinus sp.

Phellinus sp. are assigned to the Basidiomycotina, Hymenomycetes, Aphyllophorales and Hymenochaetaceae, and have been shown containing various bioactive substances including triterpenoids, polysaccharides and flavones[1]. Traditional Chinese herbalists believe that Phellinus species are effective in treating many gynecopathic ailments[2] and are also reported to exhibit other pharmacological functions including tumor cell inhibition, antioxidant activity and anti-hepatic fibrosis effects[3]. Polysaccharides of Phellinus sp. has been reported to possess antitumor activities and inhibit tumor recrudescence and metastasis. There are little studies comparing the chemical composition and biological activities difference among polysaccharides from different Phellinus sp and little report about the pure polysaccharide structure analysis. In this study, eight kinds of crude polysaccharides were extracted from Phellinus fruit bodies and their chemical composition and bioactivities were researched. The polysaccharide and protein contents of eight crude polysaccharides had a certain extent differences. Monosaccharide composition and content of amino acids also existed some differences in eight crude polysaccharides. Eight different polysaccharides all showed enhancing splenocyte proliferation effect in vitro. PB-10P and JSHP had high cell proliferation rates with 50㎍/ml concentrations. The results indicated in some extent the immune activity of crude polysaccharides were correlation with the polysaccharide and protein content and composition of each sample. The crude polysaccharides of P. igniarius were further isolated and purified using DEAE Sepharose F. F. and gel-filtration chromatography (Sephacryl S-100-500 )repeatedly. Five water-soluble homogeneous polysaccharides (P60w1、P60s1、P1SP1、P10SP1and P100SP1) were obtained. Lack of absorption at 280 nm and 260 nm by UV scanning indicated that contained no protein and nucleic acid. HPLC produced a single symmetrical peak, indicated homogeneity and their molecular weigh were 1.71×104 Da、2.07×104 Da、1.48×104 Da、2.20×104 Da and 2.56×104 Da respectively. Structural of P60w-1 were determined using sugar and methylation analysis combined with 1H and 13C NMR spectroscopy, including COSY, TOCSY, NOESY, HSQC and HMBC experiments. The effect of P60w-1 on tumor growth was examined using subcutaneously transplanted H22 and Lewis Lung Carcinoma (LLC) tumor mouse models. Cyclophosphamide or Coriolus versicolor polysaccharopeptide served as positive controls in evaluating tumor response. Results showed that P60w-1 at the most effective dose of 100 mg/kg inhibited the growth of H22 and LLC by 48% and 37%, respectively.

Antioxidant and antityrosinase properties from the fruiting bodies of Lentinus lepideus

Lentinus lepideus is an edible mushroom, belongs to the family Tricholomaceteae and order Agaricales. The purpose of this study was to evaluate the antioxidant activities, tyrosinase inhibitory effects on the fruiting bodies of L. lepideus extracted with acetone, methanol and hot water. The antioxidant activities were performed on β-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging, and ferrous chelating abilities. In addition to this, phenolic acid and flavonoids contents were also analyzed. Hot water extract of L. lepideus showed the strongest β-carotene-linoleic acid inhibition as compare to others extracts. At 8 mg/ml, methanolic extract showed a high reducing power of 1.21. The scavenging effects on 1,1-diphenyl-2-picrylhydrazyl radicals, acetonic and methanolic extracts were effective than hot water extract. The strongest chelating effect (87.50%) was obtained from the methanolic extract at 1.0 mg/ml concentration. Antioxidant activities of the extracts from the fruiting bodies of L. lepideus were increased with the increasing concentration. After application of reverse phase high performance liquid chromatography, coupled to a diode array detector and electrospray ionisation mass spectra, six phenolic compounds namely, chlorogenic acid, vanillin, naringin, naringenin, formononetin and biochanin were identified from acetonic extract. Tyrosinase inhibition of acetonic, methanolic, and hot water extracts of L. lepideus were increased with the increasing of concentration. Results revealed that acetonic and methanolic extract showed good, while hot water showed moderate activities of the tyrosinase inhibition at the concentration tested. This study suggests that fruiting bodies of L. lepideus can potentially be used as a readily accessible source of natural antioxidants.

To understand the saccharide contents and quantity of winter mushroom (Flammulina velutipes) according to its growth temperature, we measured the saccharide contents at different growth temperature. In our results, the saccharide of its fruiting body turned out to be mainly composed of xylose, trehalose and mannitol in all treatments. In the other hand, Ribose, myo-inositol and sucrose were detected in some treatments. The quantity of trehalose decreased as the growth temperature increased with a variation of its quantity depending on the isolates used in the experiments. In the case of xylose and mannitol, detected in all treatments, the pattern of their quantities was not possible to be profiled and the pattern might be largely depending on the isolates. However, the quantities of xylose and mannitol were largely in a direct proportion and the fluctuation of their quantities was congruent with the exception of ASI 4103, ASI 4166 and ASI 4065. The xylose quantity of ASI 4103 and ASI 4166 increased until the temperature was up to 10℃ and decreased when the temperature was above 10℃. That of ASI 4065 decreased as the temperature rose and increased when above 13℃. The mannitol quantity of ASI 4065 and ASI 4166 decreased until the temperature was up to 10℃ and increased when the temperature was above 10℃. That of ASI 4103 increased as the temperature rose and decreased when above 13℃.

Antioxidant, anti-inflammatory, and cytotoxic properties of fruiting bodies and their mycelia as sources of Cordyceps KCI 등재

Mushrooms play crucial roles as reservoirs of naturally occurring bioactive compounds. Among these, Cordyceps militaris is significant because of its well-established reputation for organoleptic excellence and positive health effects, which have led to its widespread commercialization. In contrast, the key properties of Paecilomyces variotii, an ectomycorrhizal symbiont, has received limited attention. In alignment with current research trends, the study of the mycelia and culture media of these mushrooms hold promise in identifying potential sources of valuable bioactive compounds. In the present study, we investigated C. militaris and P. variotii for their phenolic acids and sterols, assessing antioxidant capacity, anti-inflammatory effects, and anti-proliferative activity. Interestingly, P. variotii mycelia exhibited higher concentrations of ergosterol and phenolic compounds, with comparable levels observed in the fruiting bodies, along with superior antioxidant activity compared to that of C. militaris. In contrast, C. militaris mycelia demonstrated anti-inflammatory effects (which were absent in P. variotii mycelia) and cytotoxicity comparable to, and at times exceeding, that of its fruiting bodies (in contrast to P. variotii). In addition, the species analyzed in this study displayed variations in growth rates and mycelial production, which merit consideration for potential future applications and further study.

Protein is an essential nutrient for human health, functioning not only as an energy source but also in tissue formation and the activity of hormones and enzymes. Mushrooms are rich in β-glucans, which can enhance macrophage activity and help reduce blood cholesterol and glucose levels. Laetiporus cremeiporus, a polypore of the family Laetiporaceae, typically grows on coniferous trees and is easily recognized by its vivid yellow–orange color. This study evaluated the functional components of the mycelium and fruiting bodies of L. cremeiporus. Five strains (76, 1790, 2277, 2327, and 5379) were analyzed in this study. Experimental materials included primordia of strain 1790 grown on Quercus acutissima sawdust, mycelia cultured in potato dextrose broth (PDB), and fruiting bodies cultivated on cut logs. Protein analysis showed that the 1790 primordia had the highest content (15.2%). In strain 2327, mycelial protein (13.4%) exceeded that of the fruiting body (10.9%). β-glucan analysis indicated that 2327 mycelia had the highest content (46.4 g/100 g), while 5379 fruiting bodies contained the most among fruiting samples (33.8 g/100 g). These results demonstrate that L. cremeiporus accumulates substantial protein and β-glucan even at the mycelial stage, suggesting its potential as a functional food ingredient without requiring fruiting body production. Strain selection and cultivation optimization are essential to maximize these bioactive components.

Comparison of the metabolic profile of the mycelia and fruiting bodies of artificially cultured Cordyceps militaris KCI 등재

Cordyceps militaris, a well-known traditional Chinese medicine, has multiple health-promoting effects. It is used as a herbal remedy and health food in Asian countries. Cultured mycelia are often used as a substitute for natural C. militaris. In the present study, the mycelia and fruiting bodies of artificially cultured C. militaris were analyzed using a metabolomics approach. The protein and crude fat contents of the mycelia were substantially higher than those of the fruiting bodies. The top three abundant amino acids in the mycelia were proline (3.9 g/100 g), aspartic acid (2.9 g/100 g), and glutamic acid (2.7 g/100 g). The carbohydrate content was similar in the fruiting bodies and mycelia. Analysis revealed that both the fruiting bodies and mycelia are rich in phenolic compounds and exhibit antioxidant activity. Further, six metabolites were significantly different between the mycelia and fruiting bodies. The levels of Ca, glucose, Mg, and Se were higher in the mycelia than in the fruiting bodies. In contrast, mannitol and Zn were more abundant in the fruiting bodies. The current study provides a comprehensive metabolic profile of the mycelia and fruiting bodies of artificially cultured C. militaris. Such an exercise is potentially important for understanding the metabolism of C. militaris and facilitating the use of cultured mycelia as a supplement to C. militaris fruiting bodies in traditional Chinese medicine.
