Identification of the Hub Genes Associated with Sarcoma Through Integrative Analysis of TCGA and GEO Data KCI 등재

Background: Sarcomas are rare mesenchymal malignancies originating from connective tissues and are generally associated with poor prognosis. Previous bioinformatics studies have often relied on a single database, limiting generalizability. This study aimed to identify novel hub genes associated with sarcoma using integrative analysis of GEO (Gene Expression Omnibus) and TCGA (The Cancer Genome Atlas) datasets. Methods: Differential gene expression (DEG) analysis was performed using integrated data from TCGA and GEO. Functional enrichment analyses and protein–protein interaction (PPI) network construction were conducted, and hub genes were identified based on node connectivity. Survival analysis was performed using the Kaplan–Meier method. Results: After identifying 47 overlapping DEGs from the analysis of 261 TCGA samples and 149 GEO samples, the GO (Gene Ontology) enrichment analysis revealed associations with cell adhesion, plasma membrane components, and calcium ion binding. Moreover, the KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway enrichment analysis showed that target genes were mainly involved in the chemical carcinogenesis-receptor activation. Then, 29 genes were screened through the PPI network. With calculating protein nodes, eight genes (BCL2, EMCN, CLDN5, LYVE1, CD36, CD93, VWF, and CDH5) were screened from TCGA and GEO datasets. Comparing survival analysis outcomes among these eight genes, highly expressed CD36 (HR=0.627, log-rank p value=0.0202) was identified as being associated with improved survival outcomes in sarcoma patients. Conclusions: CD36 may serve as a candidate prognostic biomarker or survival-associated hub gene in patients with sarcoma. However, further validation is required to clarify its clinical relevance.

Brachial Plexopathy due to Myeloid Sarcoma in a Patient With Acute Myeloid Leukemia After Allogenic Peripheral Blood Stem Cell Transplantation

Myeloid sarcoma is a solid, extramedullary tumor comprising of immature myeloid cells. It may occur in any organ; however, the invasion of peripheral nervous system is rare. Herein, we report the case of myeloid sarcoma on the brachial plexus. A 37-year-old woman with acute myelogenous leukemia achieved complete remission after chemotherapy. One year later, she presented right shoulder pain, progressive weakness in the right upper extremity and hypesthesia. Based on magnetic resonance images (MRI) and electrophysiologic study, a provisional diagnosis of brachial plexus neuritis was done and hence steroid pulse therapy was carried out. Three months later the patient presented epigastric pain. After upper gastrointestinal endoscopy, myeloid sarcoma of gastrointestinal tract was confirmed pathologically. Moreover, 18-fluoride fluorodeoxyglucose positron emission tomography showed a fusiform shaped mass lesion at the brachial plexus overlapping with previous high signal lesion on the MRI. Therefore, we concluded the final diagnosis as brachial plexopathy due to myeloid sarcoma.

A hot water extract(HWE-P4) was separated from the fruit bodies of PMO-P4, and its antitumor and immunomodulatory activities against sarcoma-180 in ICR mice were investigated. The internal transcribed spacer (ITS) regions from PMO-P4 were amplified using polymerase chain reaction(PCR) and sequenced. The results revealed that PMO-P4 was belong to the Phellinus baumii. When oral administration at the dose of 160mg/kg/day in the mice until the end of the experiment with 2 week's pre-feeding of the HWE-P4, the survival rate of the mice was 152% for 50days after the inoculation of sarcoma-180 and the suppression rate of the tumor growth was 35.3%(p<0.05) for 28 days after inoculation of sarcoma-180. The HWE-P4 increased 71.4% of the CD4/CD8 ratio and 5-fold of the expression of CD25(IL- 2 receptor chain) compared with the control. From these results, the antitumor activity of HWE-P4 is exerted through its immunomodulating activity on the host's immune system.

Extraskeletal Ewing’s sarcoma (EES) is a rare tumor that may arise from soft tissues at any site. Clinically, patients usually present with painful growing mass around paravertebral region or lower extremities, and focal neurological symptoms are uncommonly accompanied. We experienced a case of an EES arising in the pelvic cavity of a 13 year-old boy who presented with pain and progressive weakness of left lower extremity for 5 months. Diagnosis of left lumbosacral plexopathy due to EES was made by electrodiagnostic examination, radiologic findings, histologic analysis, and immunohistochemical staining. Extended surgical excision and multiagent chemotherapy were performed.

Metallothionein (MT)는 세포내에 존재하는 cystein이 풍부한 적은 단백질로서 핵의학적 영상이나 치료에 이응이 가능할 여러 금속성 방사성 동위원소와 결합한다. 본 연구는 sarcoma를 주사한 Balb/C mice에서 MT와 결합하는 방사성동위원소인 cadmium이 종양에 축적되는가를 확인하고, 그것이 핵의학적인 영상과 치료에 이용될 수 있는지와 Ga-67을 대신할 수 있는지를 보기 위해 종양과 정상조직에서 Cd-109과 Ga-67의 섭취율을 비교하고 종양과 정상조직의 비를 비교하였다. Balb/3T3 세포를 Molony murine sarcoma virus (MMSV)로 변형시킨 세포를 Balb/C mice의 왼쪽 서혜부에 피하로 주사한 후 종양이 $0.6{\sim}l.0cm^2$로 자랐을 때 $25{\mu}Ci$ Cd-109 chloride와 $40{\mu}Ci$ Ga-67 citrate을 피하주사 한 후 18 또는 72시간에 쥐를 희생하여 종양과 여러 정상조직을 제거하여 무게를 잰 후 방사능을 측정하였다. Cd-109의 종양섭취는 Ga-67과 비슷하나 대부분의 정상조직에 비해 (MT가 풍부한 간과 신장을 제외) Ga-67보다 높았다. 종양대 정상조직의 섭취비는 Cd-109이 Ga-67에 비해 배후방사능에 중요한 영향을 미치는 장기인 뼈, 장, 지방조직, 근육, 혈액에서 월등히 높아서 (P<0.001) 같은 종양에서 Ga-67보다 좋은 영상을 얻을 수 있으리라고 생각되며, MT에 결합하는 cadmium은 암의 종양영상과 치료에 이용될 수 있을 것으로 생각된다.

The search for tumor-avid agents for use in nuclear medicine imaging or therapy is a field of ongoing importance. Metallothionein (MT) is an intracellular protein that binds many metals with isotopes having imaging or radiotherapeutic potential. The purpose of the study was to determine whether uptake of radioisotopes that bind to MT is increased in tumor. We measured the uptake of Cd-109 and Ga-67 in tumor and normal tissues of sarcoma-bearing mice. Tumors were grown subcutaneously in female Balb/C mice from cultured Balb/3T3 cells transformed by the Moloney murine sarcoma virus (MMSV). When the tumors reached about 1 cm in diameter, mice were injected subcutaneously with Cd-109 and Ga-67. Eighteen and seventy-two hours later, the mice were sacrified. Organs and tissues were removed, weighed, and activity per mg tissue determined by gamma well-counting. Uptake of Cd-109 by MMSV tumors exceeded that by normal tissues examined, with the exception of liver and kidney (the organs known to be richest in MT). The tumor-to-tissue ratios of uptake for Cd-109 were far greater than those for Ga-67 for many normal tissues of great importance in terms of background activity (bone, intestine, fat, muscle, and blood). We concluded that metals that bind to MT may be useful for oncologic imaging or rediotherapy of cancer.

Sarcoma 180 유발후(誘發後)의 생쥐의 생존(生存) 시간(時間)에 대(對)한 Cyclophosphamide, Corynebacterium Parvum 및 Tubercin-3의 단독(單獨) 및 병합역여(倂合役與)의 영향(影響)

Eighty of Sarcoma 180 bearing mice, averaging 30 gm of body weight, were divided into eight groups of animals receiving Saline as the control, Corynebacterium parvum, Tubercin-3 and Cyclophosphamide alone and Cyclophosphamide combined with C. parvum, with Tubercin-3 and with both C. parvum and Tubercin-3 and Tubercin-3 combined with C. parvum respectively. Treatment was initiated 4.8 hours after tumor implantation and repeated three times once a day. Doses were suspended or dissolved in 0.2 ml of Saline: 1.4 mg of C. parvum: 0.5 micrograms of Tubercin-3; and 2.7 mg of Cyclophosphamide either in alone or in combination. All the agents given were administered subcutaneously but Cyclophosphamide was given intraperitoneally. The observation on the general conditions of animal took place twice a day following the treatment until the time of death after tumor implantation was determined. Average survival days in each group were as follows: In Control, Saline (11.2 days), C. parvum (14.8 days), Tubercin-3 (16.7 days), Cyclophosphamide(18.7 days). In combination therapy, Cyclophosphamide with C. parvum(22.8 days) with Tubercin-3 (26.9 days). Cyclophosphamide with both C. parvum an Tubercin-3, however, was somewhat longer than in Cyclophosphamide alone but shorter than in combined with either one of C. parvum or Tubercin-3. Finally, in combination with immunotherapeutic agents, Tubercin-3 and C. parvum each other it (8.2 days) was shorter even than Control. Life span of host is, in generally, inversely related to the number of malignant cells and conclusively, the therapeutic potentiation was reflected to be extended survival in combined treatment of a chemotherapeutic Cyclophosphamide with either one of immunotherapeutics, Tubercin-3 or C. parvum. Tubercin-3 and C. parvum in combination, however, appeared to be antagonistic each other.

Sarcoma 180 복수암 세포에서 histone의 여러분획 즉 f1, f2a1, f2a2, f2b 및 f3로 분리 정제하여 in vitro에서 acetylation 및 methylation에 관한 연구를 행하였다. Methyl화 및 acetyl화의 관찰은 Sarcoma 180 복수암 세포의 핵을 L-methionine-Methyl-$^{14}C$ 또는 sodium acetate-1-$^{14}C$ 존재하에 jn vitro에서 행하였으며 histone 분획을 f1, f2a1, f2a2, f2b, 및 f3 별로 각각 추출 정제하였다. Methyl화 된 histone 분획들을 산으로 가수분해한 후 amino acid analyzer로 각 ammo 산을 각각 분리 정제하여 methyl화 또는 acetyl화한 amino 산 유도체 들을 동정 및 정량하였다. Histone을 methyl화 시킨 결과 polypeptide chain의 histidine 및 lysine 잔기의 유리 $\epsilon$-amino group에 methyl근이 transfer 되었다. 각 histone 분획의 비활성은 f2a1이 가장 높았으며 f2b, f2a2, f3 및 f1의 순으로 방사능이 낮었다. $\epsilon$-N-MethylIysine의 방사능을 histone의 여러 분획 즉 f1, f2a1, f2a2 및 f2b에서 증명하였으며 $\epsilon$-N-dimethyllysine의 방사능이 $\epsilon$-N-monomethyllysine 보다 1.7배나 많었다. Histone 분획 f2a2와 f2b에서는 3-methylhistidine에서도 방사능을 계측할 수 있었다. Whole histone의 각 분획에서 계측된 방사능의 값은 그 분획에서 분리한 $\epsilon$-N-monomethyllysine, $\epsilon$-N-dimethyllysine 및 3-methylhistidine의 방사능의 합계와 일치하였다. Acetyl 화의 경우 각 histone의 분획들을 trypsine과 Pronase로 가수분해하여 얻은 peptide와 amino 산들을 exclusion chromatography 및 ion exchange chromatography로 분리하였다. Acetyl 화를 행하였을 때는 arginine-rich 분획인 f2a1, f2a2 및 f3에만 incorporate 되었다. Histone의 각 분획중 f2a1이 다른 분획에 비하여 방사능이 가장 높았으며 f2a2, f3, f2b 및 f1의 순으로 방사능이 낮어진다. 각 분획의 방사능은 그의 전부가 $\epsilon$-N-acetyllysine 및 ${\alpha}$-N-acetyllysine이 갖인 방사능에 기인하였다. Lysine-rich histone 분획인 f1과 f2b에는 acetylation이 일어나지 않았으며 따라서 $\epsilon$-N-acetyllysine 및 $\alpha$-N-acetyllysine을 검출할 수 없었다.

In order to observe the properties of histones of Sarcoma 180 ascites tumor cells, the various histone fractions isolated from the tumor cells following the methylation and acetylation were studied. Histone fractions f1, f2a1, f2a2, f2b and f3 were prepared from isolated nuclei which had been incubated in the presence of L-methionine-methyl-$^{14}C$ and sodium acetate-1-$^{14}C$, respectively. For the methylation, the various histone fractions were hydrolyzed in acid and the hydrolysates were chromatographed on an amino acid analyzer. Histones were methylated by the transfer of methyl groups to the free $\epsilon$-amino groups of lysine and histidine residues in the polypeptide chain, after the histone molecules had been synthesized. The specific activity of the histone fraction f2a1 was greater than those of other fractions and the specific activities diminished in the order of f2b, f2a2, f3 and f1 fractions. Histone fractions f1, f2a1, f2a2 and f2b were found to contain radioactive $\epsilon$-N-methyllysine. $\epsilon$-N-Dimethyllysine was the predominant form of the modified amino acid and its concentration exceeds that of the $\epsilon$-N-monomethyllysine by a factor of 1.7. Histone fractions f2a2 and f2b were found to contain radioactive 3-methylhistidine. Virtually all of the radioactivity in these histone fractions are recoverable from the $\epsilon$-N-monomethyllysine, $\epsilon$-N-dimethyllysine and 3-methylhistidine. For the acetylation, each of the histone fractions was digested with trypsin and Pronase, and the resulting peptides and amino acids were separated by exclusion chromatography and ion exchange chromatography. Only the arginine-rich fractions f2a1, f2a2 and f3 were found to be appreciably labeled with acetate-$^{14}C$. Among the histone fractions, f2a1 fraction had a considerably higher specific activity than the other histone fractions and the specific activities diminished in the order of f2a2, f3, f2b and f1 fractions. All of their radioactivities could be recovered as two major radioactive peaks which were identified as $\epsilon$-N-acetyllysine and $\alpha$-N-acetyllysine. The comparatively lysine-rich histone fractions f1 and f2b were unlabeled and did contain $\epsilon$-N-acetyllysine and $\alpha$-N-acetyllysine.

전신성 홍반성 낭창 환자에서 상악 치은에 발생한 Kaposi's Sarcoma

본 증례는 전신성 홍반성 낭창으로 인해 2000년 4월부터 면역 억제제인 azathioprine 75mg과 prednisolne 35mg을 복용 중인 42세 여성 환자가 2004년 12월 상악 협측 치은의 종창, 증식, 동통 및 출혈을 주소로 내원하여 조직 생검 결과 모세 혈관종으로 진단되었으나, 2005년 1월 치은의 재발 병소의 절제 및 조직 생검으로 카포시 육종으로 진단되었고, 흉부의 피부 병변의 조직 생검과 흉부 전산화 단층 촬영 검사에서도 양측 폐에 다수의 결절을 보이는 카포시 육종으로 진단되었다. 의인성 면역억제제에 의한 카포시 육종으로 최종 진단되어, 면역 억제제와 부신피질호르몬제의 중단과, paclitaxel을 이용한 전신적인 항암화학 요법으로, 2008년11월 현재까지 재발의 징후 없이 구강, 피부, 폐의 병소에 대해 양호한 치료 결과를 얻었으나, 향후 전신성 홍반성 낭창의 악화 또는 카포시 육종의 재발 가능성이 있으므로 지속적인 추적 관찰이 요할 것으로 사료된다.

Kaposi's sarcoma was first descrided by Kaposi in 1872 as an idiopathic multiple hemorrhagic sarcoma. Its clinical features revealed to be erythematous red or purple macule started out, and developing into palpable dome-shaped nodules. Etiology is not defined to detail at present. Kaposi's sarcoma is classified to 4 categories; Classical, African, Epidemic and Immunosuppressive type. Epidemic categories is found approximately 20% of all AIDS patients and has strong predilection for head and neck region. The first case of immunosuppresive type Kaposi's sarcoma in patients with kidney transplants was reported in 1969. Kaposi's sarcoma accounts for 5% of all tumors associated with transplanted patients. The most common site of Kaposi's sarcoma in immunosuppressed patients are extremities, but rare in head and neck area. A 42 years old woman who had systemic lupus erythematosus visited to our clinic because of gingival hyperplasia, and excisional biopsy revealed Kaposi's sarcoma. We experienced a case of favorable results using excision and chemotherapy, so we report with review of literatures.

마우스에서 Sarcoma-180에 대한 Bifidobacterium adolesentis ATCC-15703의 항암 효과

Antitumor and immunoadjuvant activities of Bifidobacterium adolesentis ATCC-15703 were investigated against the ascites and solid forms of tumor induced by Sarcoma 180(S-180) in ICR mice. When 100 $\mu$g of the bacterium was intraperitonealy administered in the mice, the survival rate(T/C %) of the mice was 132% for 50 days after the inoculation of S-180, however the increment of their body weights was suppressed. When the 100 $\mu$g was subcutaneously adminis- tratered at the right grion, the suppresion rate of the tumor growth on the 21st day after inoculation of S-180 were 62.3%. Complete supression of the tumor growth was observed from 2 out of 8 test mice. The bacterium itself appeared to have noncytotoxic substance against the S-180 in vitro. The bacteria] administration markedly enhanced the activity of peritoneal macrophages. The num- ber of peritoneal macrophages at one day after the administration of the 100 $\mu$g increased about 8 times as much as that of the control, and the highest activity of acid phosphatase appeared at 2 days.

Antitumor activity of Lactobacillus casei YIP 9018(LC9018) was studied in mice by using sarcoma 180(S-180) and Lewis lung carcinoma (3LL). Following the eatablishment of in vivo tumor models such as ascites form S-180, solid form S-180 and 3LL for estimating antitumor activity of Lactobacilli, optimal dose and injection route of heat-killed LC9018 for supperssion of local tumor were examined. Administration of 100ng/mouse of LC9018 significantly inhibited the growth of ascites form S-180, solid form S-180 and 3LL.

The author observed a case of osteogenic sarcoma in a 11-year-old female with complaint of painful swelling on face in right side. The observed results were as follows: 1. Large hematoma was obsered, and patient complanited painfull swelling on c/c site. 2. Predisposing factor of osteogenic sarcoma was not clear, but patient had history of extraction before patient visiting infirmary of our dental collage. 3. Serologic findings were not specific, and serum alkaline level was normal. 4. Radiographic findings were as follows: ① Diffuse faint radiopacity in the lesion ② Bony destruction and increased radiopaciy in right antrum ③ Displacement of multiple teeth on involved area(i. e ;#12, 15. 55, 16, 17, 18), ④, Increased periodontal space in singel tooth(#13) ⑤Destruction of bony crypt on involved teeth(#13, 14, 15, 17, 18) ⑥ Loss of lamina dura of three teeth in involved area(#11, 12, 16) 5. Computed tomographic findings were as follows: ① Large calcific and hetergenous component mass in the Rt. maxillary sinus, and this mass extending to Rt. maxilla, alveolar bone, ethmoid sinus. ②, Soft tissue bulging into Rt. side nasal cavity and oral cavity. ③ Bone destruction of maxillary sinus wall and Rt. alveolar bone.

방선균 분리주 GMT 1153로부터 ts/NRK 세포의 형태를 변형된 세포형태에서 정상세포의 형태로 전환하는 활성물질을 분리, 정제하였다. 활성물질 MT 1155는 UV, IR, 1H-NMR, 13C-NMR, mass,원소분석 등의 기기분석을 통하여 항진균성 항암물질인 toyocamycin으로 동정되었다. MT 1155는 ts/NRK 세포의 형태전환에 대한 활성외에도 PKA 효소활성 저해와 CTLL 세포에 대한 세포독성이 있으나 K562 소포형성 억제 및 PKC 효소활성에 대한 저해효과도 없었다.

We isolated Actinomycetes strain GMT 1155 and purified the active compound, MT 1155, on the morphological reversion of ts/NRK cell from the isolate. MT 1155 was identified as toyocamycin having antifungal and antitumor activities from physico-chemical properties and UV, IR, $^1H$-NMR, $^13C$-NMR and mass spectrum. MT 1155 showed the morphologically reversional activity on ts/NRK cell and the cytotoxicity on CTLL cell at the final concentrations of 1.7 JlM and 0.2 11M, respectively and its $IC_{50}$ value on protein kinase A enzyme was 2.3 $\mu$M. Also it had strong antifungal activity against several pathogenic fungi but not antibacterial activity. And it did not inhibit both protein kinase C activity and the bleb-formation of K562 cell induced by phorbol esters.

1987년 3월부터 1992년 3월까지 연세의료원 암쎈터 치료방사선과에서 방사선치료를 받은 Granulocytic sarcoma 환자를 대상으로 이들의 치료성적과 방사선 선량반응 관계를 알아보기 위해 후향성 분석을 하였다. 14예의 Granulocytic sarcoma 병변이 9예의 환자에서 발생하였고 급성, 만성 골수구성 백혈병 또는 무백헐병 상태 등과 같은 다양한 임상적 시기에 발생하였다. 병소부위는 내림 순으로 골, 임파절, 연부 조직과 피부이었다. 모든 14예의 병변은 외부 방사선(Co-60 또는 전자선)으로 치료 받았고 대다수의 병변이 2000 cGy 이상의 전 방사선 선량으로 국소 방사선 치료를 받았다. 연령 분포와 발생한 임상시기는 치료에 대한 반응과 관련이 없는 것으로 나타났다. 골에서 발생한 임상시기는 치료에 대한 반응과 관련이 없는 것으로 나타났다. 골에서 발생한 병변은 다른 부위에서 발생한 병변에 비해 치료에 대한 반응이 더 좋은 것 같았다. 2000 cGy이상의 방사선 선량을 받은 병변은 $100\%$ (l1/11)의 월등한 국소 제어율을 보였지만 2000 cGy이하의 방사선 선량을 받은 병변은 $33\%$ (1/3)의 감소된 국소 제어율을 보였다. 결론적으로 국소 방사선 치료는 Granulocytic sarcoma의 완화 또는 완치 목적의 치료에 있어 효과적이면 이때 2000 cGy이상의 방사선 선량이 필요한 것으로 생각된다.

We analyzed retrospectively the patients of granulocytic sarcoma treated with radiotherapy at the Department of Radiation Oncology, Yonsei University College of Medicine from Mar 1987 to Mar.1992 in an attempt to review our experience with irradiation of granulocytic sarcoma and to evaluate the treatment results for the radiation dose response. Fourteen lesions of granulocytic sarcoma in 9 patients were developed in variable clinical settings such as AML, CML and without leukemia. The involved lesions were bone, lymph node, soft tissue and skin in descending order of occurrence. All of the lesions in 9 patients were treated with external beam radiotherapy (Co-60 or electron beam). Both age distribution and clinical settings did not show any correlation with the response to treatment. The response to treatment seemed to be better for lesions in the bone than in other involved lesions. The majority received local irradiation of a total dose of more than 2000 cGy. Radiation dose of more than 2000 cGy showed excellent local control of $100\%$, (11/11), while local control decreased to $33\%$(1/3) with total dose less than 2000 cGy. In conclusion, local radiotherapy seems to be very effective for palliative or curative aim of granulocytic sarcoma, and a radiation dose more than 2000 cGy is highly recommended.

Iridoid 계열 물질의 하나인 오쿠빈을 식나무(A. japonica)로 부터 분리한 후, 이의 생물활성작용을 조사하였다. 핵산대사 중 Sarcoma 180 암세포에 미치는 영향을 실험한 결과, 암세포 배양액에 0.1 mM, 0.5 mM, 1.0 mM의 오쿠빈을 첨가할 때 1.0 mM에서 약 30%정도 RNA 생합성 억제작용을 보여 주었다. 반면에 1.0 mM의 농도에서 DNA 생합성은 거의 억제를 받지 않았다.

Effects of aucubin on DNA and RNA biosyntheses in Sarcoma 180 cells were investigate under in vitro condition. At 1.0 mM concentration, aucubin inhibited about 30% of RNA biosynthesis, whereas at same concentration it did not appear to exhibit appreciabie inhibitory effect on DNA biosynthesis.

저자들은 지난번에 Sarcoma 180 ascites tumor cells의 histone의 생합성, methyl 화, acetyl 화 및 phosphoryl 화 등을 관찰 보고한 바 있다. 이번에는 X-선 조사가 histone 및 DNA 합성에 미치는 영향을 관찰하기 위하여 glycine-1-$^{14}C$ 및 $^{32}P$-orthophosphate를 마우스에 주사하고 실험군과 대조군을 비교 검토하였다. Sarcoma 180 ascites tumor cells를 이식하여 7~9일 된 마우스에 1250 roentgen 의 X-선을 조사하고, 대조군은 sham irradiate 하였다. 24시간 후 실험군 및 대조군의 마우스의 복강에 $10{\mu}Ci$의 glycine-1-$^{14}C$ 및 $20{\mu}Ci$의 $^{32}P$-orthophosphate를 각각 주사하고 4시간 후 tumor cells의 histone 및 DNA에서 이 표지화합물의 거동을 관찰하였다. Sarcoma 180 ascites tumor cells에서 DNA는 Ogur and Rosen의 방법으로 추출하고 각 실험군의 방사능을 계수 비교하였다. Histone은 Johns, Johns and Phillips 등의 방법으로 분리하여 f1, f2a1, f2a2, f2b 및 f3 분획을 얻었다. 각 실험군의 histone 분획들은 polyacrylamide disc gel 전기영동에 의하여 단일 단백질임을 확인하였으며, 각 분획들의 방사능을 계수 비교하였다. 대조군 및 실험군의 각 histone 분획의 amino 산의 조성은 amino acid analyzer로 분리하였고, 각 amino 산의 방사능을 계수하였다. 1. Sarcoma 180 ascites tumor cells에서 얻은 histone의 각 분획의 함량은 정상동물 즉 마우스간, 흰쥐 및 소의 흉션의 그것에 비하여 큰 차이를 나타낸다. 2. 방사선을 조사한 실험군에 있어서는 $^{14}C$-glycine의 histone에의 편입이 약 14% 감소된다. $^{14}C$-glycine의 편입은 f3 분획에 가장 높으며 f2b 분획에 가장 낮다. $^{14}C$-glycine의 histone에의 편입은 주사 4시간 후에 최고에 달한다. 3. $^{14}C$C-glycine이 histone의 glycine 분획에 편입되는 율은 방사선 조사군에 있어서 약 12% 저하한다. 4. 방사선 조사군에 있어서는 DNA에 연입되는 $^{32}P$의 율이 약 14% 저하한다. Histone 생합성과 DNA 대사와의 관련에 관하여 약간의 고찰을 행하였다.

In the earlier works from this laboratory, biosynthesis, acetylation, methylation and phosphorylation of histone of transplantable ascites tumor cells have been studied. The present paper deals with the effects of X-ray irradiation on histone biosynthesis of Sarcoma 180 ascites tumor cells. The tumor-bearing mice were irradiated with a dose of 1250 roentgen prior to 24 hours double-labeling with $10\;{\mu}Ci$ of glycine-$^{14}C$ and $20\;{\mu}Ci$ of $^{32}P$-orthophosphate. Sham irradiated mice were used as control. Histones were prepared by Johns procedure and fractionated on polyacrylamide disc gel electrophoresis. The histones were hydrolyzed in acid and the digests were chromatographed on an amino acid analyzer. DNA was extracted according to the method of Ogur and Rosen. Radioactivity was counted in a liquid scintillation spectrometer. The time course changes in the incorporations of labeled compounds into histone and DNA have been followed. Results obtained were as follows; 1. Contents of histone fractions from Sarcoma 180 ascites tumor cells have been shown to markedly differ from normal tissues, i.e. mouse liver, rat and calf thymus. 2. X-ray irradiation hindered $^{14}C$-glycine incorporation into histone by 14 per cent. Uptake of $^{14}C$ was highest into f3 and was lowest into f2b among histone fractiones both in normal and irradiated mice. The incorporation of $^{14}C$-glycine into histone reached a maximum 4 hours after glycine administration. 3. Radioactivity of $^{14}C$-glycine was less distributed about 12 per cent in glycine fraction of histone hydrolysates from X-ray irradiated animal. 4. A decreased $^{32}P$-incorporation (about 14%) into DNA was observed in irradiated mice. Relationships between histone biosynthesis and DNA metabolism were also discussed.

흉쇄유돌근에서 기인한 Low-Grade Myofibroblastic Sarcoma 1예

Low-grade myofibroblastic sarcoma is a rare disorder of malignant soft tissue tumor and is usually occurred various regions that often manifests in the head and neck region. The most common clinical presentation of this entity is non-tender cervical lymphadenopathy. This disease usually presents high possibility of local recurrence and low possibility of distant metastasis. We report a rare case of low-grade myofibroblastic sarcoma occurring in the sternocleidomastoid muscle and invading to the mastoid tip. A 56-year-old male visited the clinic with a complaint of slowly growing postauricular mass for 6 months. Mass originating from sternomastoid muscle and invasion to mastoid tip was observed by imaging studies. Surgical complete excision with simple mastoidectomy was performed via modified facelift approach. The histopathologic findings revealed malignant spindle cell tumor with positive staining with smooth muscle actin. The patient has no recurrence for 2years without any complication.
