Galectins are well-known β-galactoside-binding proteins, which play vital roles in innate immune responses of both vertebrates and invertebrates. However, knowledge regarding invertebrate galectins is still in its infancy. With the intention of filling the knowledge gap, here we identified a quadruple domain-containing galectin from marine invertebrate disk abalone, Haliotis discus discus (AbGalec), and characterized it. AbGalec consisted of four distinct carbohydrate-recognition domains (CRDs) and lacked a signal peptide. Expression analysis revealed AbGalec to be ubiquitously expressed in all the examined early embryonic stages of abalone, with highest expression in the 16-cell stage, suggesting the importance of AbGalec in early developmental processes. Tissue distribution analysis revealed the highest expression of AbGalec in abalone mantle, followed by that in gills and hemocytes. Immune challenge experiments revealed significant upregulation of AbGalec at 24 h and 48 h post injection (p.i.) with bacterial and viral components. These results suggested the possible involvement of AbGalec in host defense mechanisms. Polyinosinic: polycytidylic acid (Poly I:C) and viral hemorrhagic septicemia virus (VHSV) injections were capable of inducing AbGalec transcript expression more prominently than bacterial stimulants, thus providing evidence for its role in viral infections. We determined the virus-neutralizing ability of a quadruple domain-containing galectin for the first time, by analyzing the downregulation of VHSV transcripts during the overexpression of AbGalec. Significant downregulation of VHSV transcripts was observed after 24 h and 48 h of post infection. Collectively, our findings reveal the potent antiviral responses of molluscan quadruple domain-containing galectin, AbGalec, along with its involvement in innate immunity.

Interferon regulatory factors (IRFs) are among the most important transcription mediators and have multiple biological functions, such as antiviral and antimicrobial defense, cell differentiation, immune modulation, and apoptosis. Three IRF family members (HaIRF4-like, HaIRF6, and HaIRF8) of the big belly seahorse (Hippocampus abdominalis) were molecularly and functionally characterized at the sequence and transcriptional level. The coding sequences of HaIRF4-like, HaIRF6, and HaIRF8 were 1214, 1485, and 1266 bp in length, encoding proteins of size 46.21, 55.32, and 47.56 kDa, respectively. Potential viral transcription and replication was detected against VHSV infection using qPCR in HaIRFs-transfected FHM cells. IRFs significantly reduced viral gene expression at 24 h and 48 h post infection and the expression of interferon-stimulated genes (ISGs) was modulated at transcriptional level upon HaIRF overexpression in FHM cells. Subcellular HaIRF localization was observed using GFP-tagged expression vectors in FHM cells. HaIRF4-like and HaIRF8 were localized to the nucleus, whereas HaIRF6 was observed in the cytoplasm. All three IRFs were ubiquitously expressed in all analyzed tissues of the big belly seahorse. The mRNA expression of IRF4-like, IRF6, and IRF8 increased significantly post injection in the blood and gills following LPS, poly (I:C), and Streptococcus iniae challenge. These findings demonstrate that seahorse IRFs are involved in host defense mechanisms against immune stimulants and HaIRFs induce interferon and ISGs which trigger antiviral activity against viral infections in the host.

The mRNA Expression and Methylation Pattern of Apoptosis-related and Imprinted Genes in Day 35 of Cloned Pig Fetuses

This study was conducted to examine the mRNA expression of apoptosis-related and imprinted genes and methylation pattern of the differentially methylated region (DMR) of H19 gene in day 35 of SCNT pig fetuses. The day 35 of natural mating (control) or cloned (clone) pig fetuses were recovered from uterus. Endometrium from dam and liver from fetus were obtained, respectively. mRNA expression was evaluated by real-time PCR and methylation pattern was analyzed by bisulfite sequencing method. The Bcl-2 mRNA expression in clone was significantly lower than that of control (p<0.05). The mRNA expression of H19 gene in both endometrium and liver was significantly higher in clone than that of control, respectively (p<0.05). The level of IGF-2 mRNA in liver of clone was significantly lower than that of control (p<0.05), whereas the mRNA expression of IGF2-R gene in liver of clone was significantly higher than that of control (p<0.05). The DMR of H19 was lower methylation pattern in clone than that of control. These results suggest that the aberrant mRNA expression of apoptosis-related and imprinted genes and the lower DMR methylation pattern of imprinted gene may be closely related to the inadequate fetal development of cloned fetus.

A chymotrpsin gene homologue was cloned from the mulberry longicorn beetle, Apriona germari. The A. germari chymotrypsin cDNA contains an ORF of 950 nucleotides capable of encoding a 283 amino acid polypeptide with a predicted molecular mass of 29151 Da and pI of 9.38. The A. germari chymotrypsin has conserved six cysteine residues and active triad formed by His, Asp and Ser. The deduced amino acid sequence of the A. germari chymotrypsin cDNA was closest in structure to the Anthonomus grandis chymotrypsin. Northern blot analysis revealed that A. germari chymotrypsin showed the midgut-specific expression.

Decreased Pattern-Recognition Receptor-Mediated Cytokine mRNA Expression in Obese Children With Otitis Media With Effusion

Objectives. To assess innate and humoral immune responses in middle ear effusion of obese pediatric patients with otitis media with effusion (OME). Methods. We evaluated 219 children with OME, of whom 21 were obese and 198 were non-obese. We compared the ex- pression in middle ear effusion of mRNAs encoding toll-like receptors (TLR) 2, 4, 5, and 9; nucleotide-binding oligo- merization domains (NOD) 1 and 2; retinoic acid-inducible gene (RIG)-I; interleukins (IL)-6, -10, and -12; interferon (IFN)-γ; and tumor necrosis factor (TNF)-α mRNAs. We also compared the expression of immunoglobulins IgG, IgA, and IgM and the bacterial detection rate in the two groups. Results. TLR2-mediated expression of IL-6 mRNA, TLR4-mediated expression of IL-6 and IL-10 mRNA, TLR5-mediated expression of IL-6, IL-10, and TNF-α mRNA, TLR9-mediated expression of IL-6 mRNA, and NOD2-mediated ex- pression of IL-6, IL-12, and TNF-α mRNA were significantly lower in obese than in non-obese children (P<0.05). However, concentrations of IgG, IgA, and IgM in middle ear effusion were lower in obese than in non-obese children, but none of these differences was significant (P>0.05). Conclusion. Mean body mass index was higher and pattern-recognition receptor-mediated cytokine mRNA expression was lower in obese than in non-obese children with OME.

Androgen plays an important role in the regulation of gonadotropin production in vertebrates . We have investigated the transcriptional pattern of androgen receptor (AR) in a variety of tissues in maturing male and female goldfish by RT-PCR. Specific primer for AR was designed based on goldfish AR gene from the GenBank (accession number AY090897). AR was shown 10 be maturity- and tissue-dependent gene expression pattern in goldfish. In immature male goldfish, significantly higher transcript level of AR was observed in the pituitary und testis , compared [0 brain and liver. Mature male goldfish showed a similar expression pattern to immature male goldfish. Interestingly. when compare to male goldfish, female goldfish showed AR mRNA expression that was found 10 be weak in pituitary, and very low expression in brain. They could not be found 10 have expression in any other tissues. Taken together. the- transcriptional analysis of AR depending on the tissue, sex. and maturity of a goldfish provides the opportunity for the study of goldfish reproductive physiology ,The results provided for the first time a comparison of the tissue distribution of AR mRNA in sexually maturating male and female goldfish.

Temporal Pattern of cAMP Concentrations and α-Actin mRNA Expression in Skeletal Muscle of Cimaterol-Fed Rats

Twenty four female Sprague-Dawley rats weighing about 190 g were used to examine changes in muscle cAMP concentrations and steady-state levels of skeletal muscle ${\alpha}$-actin mRNA during chronic administration of cimaterol, a ${\beta}$-adrenergic agonist. Cimaterol was mixed in a powdered rat diet at 10 mg/kg diet. At 3 and 21 days after the start of treatment, skeletal muscle and heart samples were collected for the measurement of cAMP concentrations and skeletal muscle ${\alpha}$-actin mRNA levels. Cimaterol increased (p < 0.01) body weight gain gradually during the first seven days of the trial period, but not thereafter. Most skeletal muscle weights and the ratio of muscle weight to body weight were increased (p < 0.05) by cimaterol treatment both at 3 and 21 days. Heart weight was also increased (p < 0.05) by cimaterol treatment at 3 and 21 days, but the ratio of heart weight to body weight was increased (p < 0.05) only at 3 day. Cimaterol decreased (p < 0.05) cAMP concentration of gastrocnemius muscle at both 3 and 21 days after treatment. However, cimaterol tended (p = 0.07) to increase cAMP concentration at 3 days in the heart. Cimaterol tended (p = 0.08) to increase the steady-state level of ${\alpha}$-actin mRNA by 60% in gastrocnemius muscle at 3 days but had no effect at 21 days. The results indicate that the pattern of hypertrophic response to chronic dietary administration of cimaterol is different between cardiac and skeletal muscle. In skeletal muscles it appears that the hypertrophy induced by cimaterol is partly due to stimulated myofibrillar protein synthesis at a pre-translational level.

The bio-complex "reaction pattern in vertebrate cells"(RiV) is mainly represented by characteristic exosome-like particles - probably as reaction products of cells to specific stress. The transcription factor NF-kappaB plays a central role in inflammation. We tested the hypothesis that RiV particle preparations (RiV-PP) reduce cellular adhesion molecule (CAM) expression (ICAM-1, VCAM-1, E-selectin) by the attenuation of NF-kappaB translocation in human umbilical vein endothelial cells (HUVEC). After 4 hours, pre-incubation of HUVEC with RiV-PP before stimulation with TNF-alpha significantly reduced ICAM-1 (65.5${\pm}$10.3%) and VCAM-1 (71.1${\pm}$12.3%) mRNA expression compared to TNF-alpha-treated cells (100%, n=7). ICAM-1 surface expression was significantly albeit marginally reduced in RiV/TNF-alpha- treated cells (92.0${\pm}$5.6%, n=4). No significant effect was observed on VCAM-1 surface expression. In RiV/TNF-alpha-treated cells (n=4), NF-kappaB subunits p50 (85.7${\pm}$4.1%) and p65 (85.0${\pm}$1.8%) nuclear translocation was significantly reduced. RiV-PP may exert an anti-inflammatory effect in HUVEC by reducing CAM mRNA expression via attenuation of p50 and p65 translocation.

정상자궁근조직과 자궁근종조직에서 FSTL1유전자 클로닝 및 발현양상

목적 : 정상자궁근조직과 자궁근종조직사이에서 다르게 발현되는 유전자 클로닝 및 발현양상을 비교·분석을 위한 실험을 실시하고자 한다. 연구 방법 : 광주기독병원 산부인과에서 2002년 9월부터 2003년 9월까지 자궁적출술, 근종절제술, 제왕절개술을 받은 25명의 환자의 정상자궁근조직과 자궁근종조직에서 total RNA를 분리하였다. Differential display PCR에서 분리된 유전자를 클로링하여 유전자염기서열분석을 하였으며, PCR을 이용하여 자궁근종조직과 정상자궁근조직사이에서 유전자 발현비의 양상을 비교·분석하였다. 유의치 검증은 paired t test을 이용하여 p<0.05로 하였다. 결과 : Differential display PCR에 의해 분절된 유전자는 FSTL1으로 자궁근종조직과 정상자궁근조직에서 FSTL1의 발현비의 양상을 보면, 생식연령기 여성에서는 정상자궁근조직 및 인접 정상자궁근조직에서 자궁근종조직보다 유의하게 (p<0.05) 높게 나타났으며, 이에 반하여 폐경연령기 여성에게서는 자궁근종조직에서 유의하게 (p<0.05) 높게 나타났으며, 인접 정상자궁근조직에서는 유의한 차이는 없었다. 자궁근종 축소 호르몬치료군에서 1.4-1.6배 유의하게 (p<0.05) 높았고, 폐경기 증상완화 비치료군에서 1.6-2.0배 유의하게 (p<0.05) 높았다. 결론 : FSTL1의 작용기전은 확실하지 밝히지 못했지만, 자궁근종조직과 정상자궁근조직의 발현양상을 비교·분석해 볼 때 자궁근종에서의 FSTL1은 estrogen에 의해 직·간접적으로 조절되는 자궁근종성장억제인자일 것으로 사료된다.

Objective : To study the influence and cloning of differentially expressed genes in human female normal myometrium and uterine leiomyoma tissue. Methods : In this experiment, human uterus tissues (n=25) were taken for total RNA isolation by using Trizol reagent. Differential display was performed by using GeneFishingTM DEG Kit and processed to cDNA sequencing and gene cloning for Follistatin-like 1 (FSTL1). Data were analyzed with the image Master VDS software and statistical significance was defined as p<0.05 by paired t test results. Results : FSTL1 mRNA expression level was significantly higher (p<0.05) in normal and adjacent normal myometrium tissues than uterine leiomyoma tissue of women in the reproductive age. Whereas in the menopausal age, FSTL1 mRNA expression level was significantly higher (p<0.05) in uterine leiomyoma than normal myometrium. There was no significant differences between uterine leiomyoma and adjacent normal myometrium. Conclusion : Although the mechanisms of FSTL1 gene were uncertain, FSTL1 seemed to play an important role in the growth of uterine leiomyoma, it also might be related to the regulation of uterine leiomyoma growth inhibiting factors by modulating Follistatin related protein gene (FLRG) system.
