프럭토올리고당 첨가가 반죽의 물성과 식빵의 품질 특성에 미치는 영향

This study was conducted to evaluate characteristics of bread and rheology of flour dough containing fructooligosaccharide. In the farinogram test, addition of fructooligosaccharide changed stability and development time. Stability and development time increased as the amount of fructooligosaccharide increased. In the extensograph test, the degree of extension decreased with increasing of fructooligosaccharide content, whereas degree of resistance increased. The volume of the loaf bread containing 10% fructooligosaccharide was the largest, and the bread containing 15% fructooligosaccharide revealed the best taste, flavor, and texture scores. After fermentation treatment, the volume of the dough with 20% of sucrose were less than that of 20% of fructooligosaccharide. Dough with 20% of sucrose revealed lowest dough raising power compared to other doughs. Overall, preference scores by sensory evaluation of bread containing fructooligosaccharide were higher than that of bread with sucrose. Therefore, excellent quality of bread may be achieved by adding fructooligosaccharide instead of sucrose to the process.

This study was carried out to compare the effect of dietary zinc gluconate, fructooligosaccharide and probiotic on body weight gain, feed intake and feed efficiency of broiler chicks. A feeding trial was conducted with a total of 216 broiler chicks. Each treatment had 18 chicks with 3 replications. The supplementation levels of each treatment in the experimental diets were 0.20% for zinc gluconate, fructooligosaccharide and probiotic, respectively. The body gain of chicks fed feed additives significantly increased (p<.05) compare with that of control. Especially, the treatment with probiotic produced higher body gain than other treatments. Average feed intake of chicks was not significantly different among treatments (p<.05). Feed efficiency of chick fed feed additives significantly (p<.05) improved compare with control and the highest in the probiotic treatment. Therefore, the feed efficiency of chicks fed fructooligosaccharide, zinc gluconate and probiotic treatment were increased by 3.5%, 4.0% and 4.5%, respectively compare to that of the control. In conclusion, dietary feed additives tended to slightly improve the body gain and feed efficiency. Especially the treatment with probiotic produced higher body gain and feed efficiency among other treatments.

Loperamide에 의한 변비는 위 배출, 소장 및 대장 운동성을 감소시켰으며, 과채복합추출물(Fruit and vegetable complex extracts, FVCE)은 이러한 효과를 예방한다. 우리는 다음과 같은 실험에서 로페로마이드(3 mg/kg, sc, 14일)로 유발된 변비 수컷 Sprague-Dawley 쥐에 대한 프락토올리고당 및 과채복합추출물의 효과를 조 사하였다. 쥐를 무작위로 정상대조군 쥐(일반식이), 변비 쥐(일반식이 + 로페로마이드), 200mg FVCE(200mg/kg/day FVCE + 로페로마이드가 보충된 일반 식이)로 처리한 변비 쥐, 400mg FVCE로 처리한 변비 쥐(200mg/kg/day FVCE와 로페로마이드가 보충된 일반 식단)으로 시험군을 구성하였다. 대조군 쥐에 비해 로페로마이드로 처리된 쥐 에서 대변 배설량이 적고 대변 수분 함량이 더 낮았다. FVCE을 경구투여 하면 분변배설량의 감소를 억제하고 로페 로마이드로 처리된 쥐의 대변 수분 함량. 로페로마이드를 처리한 쥐에서는 대장선와세포의 점액 생성과 대변 및 점막 표면의 점액 함량이 감소하였다. 그러나 FVCE 처리군에서는 대장선와세포의 뮤신 함량이 증가하였고, Aclain blue 로 염색된 점액층은 로페로마이드 처리군에 비해 FVCE 처리군에서 유의적으로 두꺼워졌다. 쥐 회장에서 로페로마이 드는 회장 운동성을 억제한다. 이러한 결과는 FVCE가 로페로마이드에 의한 대장 연동 운동 억제 완화에 효과적이며 FVCE추출물이 변비 예방에 효과적 일 수 있다는 것을 나타낸다.
Loperamide-induced constipation reduced gastric emptying, small-intestinal and colonic motility, and these effects were prevented by Fruit and vegetable complex extracts(FVCE). In this study, the effects of FVCE on constipated male Sprague-Dawley rats induced by loperamide(3 mg/kg, s.c.,14 days) were investigated. Rats were randomly assigned to the normal control rats(regular diet), constipated rats(regular diet plus loperamide), constipated rats treated with 200 mg FVCE(regular diet supplemented with 200 mg/kg/day FVCE plus loperamide), constipated rats treated with 400 mg FVCE(regular diet supplemented with 400 mg/kg/day FVCE plus loperamide). There was less fecal excretion and lower fecal water content in loperamide-treated rats than in control rats. Oral administration of FVCE blocked the decrease of fecal excretion and fecal water content in the loperamide-treated rats. Mucus production of crypt cell and mucus contents at fecal and mucosa surface were reduced by loperamide-treated rat. But colonic crypt cell contained increased mucin in the FVCE treated group and mucus layer stained with alcian blue was significantly thicker in FVCE treated rats compared with in loperamide-treated rats. In isolated rat ileum, loperamide produced inhibition of ileal motility. Pretreatment with water extracts of FVCE in isolated rat ileum prevented inhibition by loperamide. These findings indicated that the FVCE was effective for alleviation of inhibition of colonic peristalsis by loperamide and that FVCE might be of value in the prevention of constipation.

This study sought to investigate any additive or interactive effects that soy isoflavones may have on the ecosystem of the gut, which is influenced by fructooligosaccharide (FOS) in colon-cancer model rats. Male Sprague-Dawley rats treated with 1,2-dimethylhydrazine were given experimental diets containing 0, 3, 6, or 9% FOS with or without 0.1% soy isoflavone for 12 weeks. In addition to the effects of FOS dosage on the gut ecosystem, dietary supplementation with soy isoflavone reduced the number of colonic aberrant crypts (ACs). The fecal weight, fecal pH, and gut transit time significantly decreased in a dose-dependent manner in rats fed FOS and the fecal concentration of bifidobacteria was higher in rats fed FOS than in control rats. The fecal output of total short-chain fatty acids, acetate, and propionate was significantly increased by the presence of FOS and was negatively correlated with the number of ACs, whereas the fecal output of butyrate showed no significant correlation with FOS dosage. The addition of soy isoflavone to the diet did not result in any significant differences in gut ecosystem parameters. Therefore, we conclude that the suppressive effect of soy isoflavone on ACs was not associated with the intestinal ecosystem, which was significantly altered by the dosage of FOS.
갓김치로부터 분리한 Probiotic 유산균과 Prebiotic Fructooligosaccharide로 제조한 요구르트의 Synbiotic 가능성

갓김치로부터 분리한 probiotic L. acidophilus GK20 및 L. paracasei GK74를 단독 혹은 혼합 배양하여 제조한 요구르트를 저장하는 동안 이화학적 및 미생물학적 특성과 스타터의 위산이나 담즙산에 대한 저항성, Caco-2 cell에 대한 부착능, 항균, 항산화 및 효소적 활성에 대한 prebiotic FOS(fructooligosaccharide)의 영향을 살펴보았다. FOS를 첨가했을 때 요구르트 내의 스타터 균수, 총산도 및 점도는 유의하게 높아짐과 동시에 pH는 감소되었다(p<0.05). 또한 E. coli ATCC 11229와 S. enteritidis ATCC 13076은 probiotics 스타터가 생산한 항균물질에 의해 저해되었으며, FOS를 첨가한 synbiotic 요구르트의 항균활성은 더욱 증가되었다. 게다가 FOS (1.0%)를 첨가하여 혼합 스타터로 발효시킨 요구르트에서 가장 높은 단백질 분해능 ($1.06{\pm}0.06$ unit) 및 ${\beta}$-galactosidase 활성 ($20.14{\pm}0.31$ unit)을 나타내었다. 하지만 비록 L. acidophilus GK20과 L. paracasei GK74 모두 장관액에 대한 저항성, 장관상피세포에 대한 부착능 및 DPPH radical 소거능이나 환원력과 같은 항산화 활성을 나타내었지만, 이들 활성이 FOS에 의해 증가되진 않았다. 결과적으로 L. acidophilus GK20과 L. paracasei GK74 혼합 배양에 FOS를 첨가한 synbiotic 요구르트는 장내환경 개선과 건강기능 향상에 유용한 것으로 여겨지며, 생리활성은 $4^{\circ}C$에서 7일간 저장 하에서도 일정하게 유지되었음을 확인하였다.
In the present work, the influence of prebiotic fructooligosaccharide (FOS) on adhesion to Caco-2 cells, viability, acid and bile tolerance, antibacterial, antioxidant, enzymatic, and metabolic activities of the probiotic starters Lactobacillus acidophilus GK20 and Lactobacillus paracasei GK74, has been explored. Experiments were conducted with fermented yoghurt over a period of 7 days at $4^{\circ}C$. When compared to control fermentations without prebiotic, the addition of FOS was seen to significantly (p<0.05) increase the viable cell counts of the probiotics, overall viscosity, and concurrently reduce the pH of the fermented yoghurts. Both Escherichia coli ATCC 11229 and Salmonella enteritidis ATCC 13076 were inhibited by the probiotics' antibacterial activities, while the synbiotic yoghurt containing mixed probiotics and FOS was noted to highly improve antagonistic action. When fermented with mixed starters, the addition of FOS (1.0%) resulted in the highest proteolytic ($1.06{\pm}0.06$ unit) and ${\beta}$-galactosidase activities ($20.14{\pm}0.31$ unit). However, FOS did not affect acid and bile tolerance, adhesion to Caco-2 cells or the antioxidant activity of the probiotics, although both L. acidophilus GK20 and L. paracasei GK74 had functionality as probiotic strains. Hence, a significant synbiotic effect was observed in fermented yoghurt after 7 days of storage at $4^{\circ}C$, and as a result, such synbiotic yoghurt can be said to possess synergistic actions which improve the gastrointestinal environment and promote of health.

One hundred and twenty-eight growing barrows (Jiaxing Black${\times}$Duroc${\times}$Landrace) at an average BW of 20.8 kg were allocated to four treatments for 42 days, each of which was replicated four times with eight pigs per replicate and used to investigate the effects of fructooligosaccharide (FOS) on digestive enzyme activities, intestinal microflora and morphology of growing pigs. The pigs received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0, 2, 4, 6 g/kg diet at the expense of corn, respectively. As compared to control, supplementation with 4 and 6 g/kg FOS significantly improved average daily gain and feed efficiency. Addition of FOS enhanced the growth of Bifidobacterium and Lactobacillus, but inhibited Clostridium and Escherichia coli in the small intestinal and proximal colonic contents. Supplementation with 4 and 6 g/kg FOS significantly improved the activities of total protease, trypsin and amylase in the small intestinal contents. However, FOS had no significant effect on the activity of lipase in the small intestinal contents as well as the digestive enzymes in pancreas. Morphological measurement of jejunal mucosa did show response to consumption of FOS. Villus height and the villus height to crypt depth ratio at the jejunal mucosa were significantly higher with 4 and 6 g/kg FOS supplementation as compared to control.

This study was carried out to investigate the efficacy of sn-2 palmitic acid-fortified vegetable oil (Sn2PA) on calcium absorption and to confirm the synergistic effects of fructooligosaccharide on calcium absorption. Male SD rats were fed 6 kinds of casein based diets containing vegetable oil (control), sn-2 palmitic acid-fortified vegetable oil (Sn2PA) and Sn2PA with fructooligosaccharide(Sn2PAFO) in two levels of calcium (normal 0.5% and high 1.0%) for 3 weeks. Total lipids, cholesterol, triglyceride and calcium in blood were measured. Feces were collected using cages for 4 days. Serum concentrations of total lipids and calcium were not significantly different among groups. However, serum triglyceride was significantly decreased by fructooligosaccharide supplementation regardless of dietary calcium level. The lipid absorption was not significantly different among experimental groups. Calcium absorption was significantly higher in Sn2PAFO group than other groups. Calcium solubility of intestine was increased by sn-2 palmitic acid supplementation. These results suggest that sn-2 palmitic acid and fructooligosaccharide supplementation could be beneficial for baby foods including infant formula, with regard to increasing absorption of calcium by more soluble calcium in the small intestinal content.

We investigated the combinatorial effects of different doses of dietary soy isoflavones (SI) and fructooligosaccharide (FOS) in a rat model of colon cancer. We hypothesized that increased bioavailability of SI metabolites due to dietary FOS may increase production of bioactive equol and affect colon carcinogenesis in a dose-dependent manner. Sprague-Dawley male rats were injected with 12-dimethylhydrazine (DMH) and were providec experimental diets that contained 0, 10, 50, 150, or 500 mg SI per kg of diet and 6% FOS for 12 weeks. The number of aberrant crypt foci (ACF) and the expression of cyclooxygenase-2 (COX-2) in colonic tissues were significantly decreased in the 6% FOS-fed groups compared to the control group. Gut transit time and fecal pH were significantly lower, and fecal concentrations of bifidobacteria were increased with 6% FOS. However, dietary SI supplementation in combination with 6% dietary FOS did not affect ACF formation or COX-2 expression. Plasma equol concentrations were dose-dependently increased by supplementation of SI up to 500 mg/kg of diet. In conclusion, SI supplementation up to 500 mg/kg of diet appeared to have no additive beneficial effects in rats with chemically-induced colon cancer that were fed 6% FOS, although plasma equol was dose-dependently increased.
프락토올리고당 첨가 블루베리청의 이화학적 품질 특성에 관한 연구

본 연구에서는 기존에 사용하고 있는 고열량 당분인 설탕 대신에 프락토올리고당을 사용하여 블루베리 청을 제조하고자 하였다. 설탕 함량, pH 및 색도 측정을 하였고, 항산화활성 및 페놀성 화합물의 함량을 측정하여 생리활성을 평가하였다. $L^{\ast}$ 값(명도), pH 및 색상은 프락토올리고당의 배합 비율이 높은 청에서 높았다. $a^{\ast}$값(적색도)은 열처리된 청에 대해 높았으며, $b^{\ast}$ 값(황색도)은 $L^{\ast}$ 값과 반대되는 경향이 있었다. 또한 열처리된 프락토올리고당 함유 청은 열처리 하지 않은 청보다 총 폴리페놀, 플라보노이드 및 안토시아닌의 함량이 더 높았다. 블루베리 청의 항산화 활성은 페놀성 화합물의 함량에 따라 차이를 보였다. 조분쇄 생블루베리와 프락토올리고당을 1:1 비율로 배합하여 열처리한 S2 샘플이 가장 높은 값을 나타내었다. 이러한 결과는 사용된 감미료의 맛에 대한 우리의 친숙함이 우리의 취향에 영향을 미친다는 것을 시사한다. 따라서 설탕에 적합한 대안을 찾기 위해 프락토올리고당을 포함한 새로 개발된 감미료를 연구할 필요가 있다.
The purpose of this study was to prepare blueberry syrup (cheung) using fructooligosaccharide instead of the traditionally used sucrose. The sugar content, pH, and colorimetric values were measured to evaluate the quality of the prepared syrup, and the antioxidant activity and phenolic compound content were determined to assess the physiological activity of the syrup. The $L^{\ast}$ value (lightness), pH, and color were high in syrups containing a high proportion of fructooligosaccharide. The $a^{\ast}$ value (redness) was high in syrups subjected to heat treatment, and the $b^{\ast}$ value (yellowness) tended to be the opposite of the $L^{\ast}$ value. Moreover, syrups containing heat-treated fructooligosaccharide showed higher polyphenol, flavonoid, and anthocyanin contents than the unheated syrups did. The antioxidant activity of the blueberry cheung depended on the content of phenolic compounds. The highest value was obtained in the cheung prepared using a heat-treated 1:1 mixture of blended raw blueberry and fructooligosaccharide (S2). These findings suggest that our familiarity with the taste of the sweetener used influences our taste preference. It is therefore necessary to study other newly developed sweeteners apart from fructooligosaccharide to find a suitable alternative to sucrose.
차전자피 분말을 첨가한 프락토올리고당 딸기잼의 관능적 특성

This study investigated the sensory characteristics of strawberry jam sweetened with fructooligosaccharide added with psyllium husk powder: 7 groups CON (0%), SSP2 (2%), SSP4 (4%), SSP6 (6%), SFP2 (2%), SFP4 (4%), SFP (6%). The moisture content of strawberry jam did not show significant difference by adding psyllium husk powder while samples sweetened with fructooligosaccharide(SFP) showed a significantly higher moisture content than samples sweetened with sugar(SSP). The pH value showed no significant difference by adding psyllium husk power and fructooligosaccharide. ˚Brix did not make significant difference by adding psyllium husk powder, but SSP showed higer ˚Brix than SFP. In terms of color, brightness (L) and redness (a) tended to decrease as psyllium husk powder were added, while yellowness (b) showed just opposite propensity. Spreadability increased in the jam added by psyllium husk powder. Attribute difference test resulted that the score of jelly appearance, sticking, soil odor, oriental medicine odor, oriental medicine flavor, bitterness, chewiness, jelly texture, roughness were evaluated to be the highest in SFP6 while gloss, strawberry flavor, sweetness, sourness, moistness indicated highest in CON. Consumr acceptance acceptance results showed SFP2 had the highest in samples. Limitations and future research directions of this study also discussed.

본 연구는 프락토올리고당(95%)과 변성전분 AA의 혼합사용 떡의 노화 억제 효과 분석을 위하여 중심합성계획법에 따라 쌀가루 기준으로 프락토올리고당(95%)은 0, 3, 6, 9 또는 12% 농도로 첨가하고, AA 전분은 0, 0.3, 0.6, 0.9 또는 1.2% 농도로 혼합사용하여 제조한 떡을 5℃에서 냉장 보관하면서 제조시간 2시간과 24시간 경과 후 기계적 텍스처 특성과 관능적 묘사 특성을 분석하였다. 기계적 텍스처 변화에서 제조 2시간 후 탄성, 응집성, 씹힘성, 검성, 부착성 및 경도 등 모든 텍스처 특성에서 각 실험군 간의 차이가 있었다(p<0.001). 제조 24시간 후 탄성을 제외한 응집성, 씹힘성, 검성, 부착성 및 경도 등에서 각 실험군 간의 차이가 있었고(p<0.001), 프락토올리고당(95%) 6%와 AA 전분 1.2% 수준으로 혼합 첨가하면 텍스처 경도가 가장 낮게 측정되었다. 관능적 특성에서는 제조 2시간과 24시간 후 AA 전분의 함량이 높을수록 경도는 감소하고, 프락토올리고당(95%)의 함량이 높을수록 단맛은 증가하는 경향을 보여주었으며, 가래떡 제조 24시간 후 프락토올리고당(95%) 6%와 AA 전분 1.2% 수준으로 혼합 첨가하면 경도가 가장 낮게 조사되었다. 텍스처 변화와 관능적 특성 간의 상관관계를 분석 결과 모든 기계적 텍스처 특성은 관능적 특성 중 부착성, 경도 및 응집성과 높은 상관관계를 나타내었다(p<0.001). 따라서 프락토올리고당(95%)과 변성전분 AA의 혼합사용의 경우 프락토올리고당(95%) 6%와 AA 전분 1.2% 농도 수준으로 혼합 첨가하면 저장 중 떡의 노화 억제에 효과가 있는 것으로 나타났다.
The quality characteristics of Korean rice cakes (Karedduk) with a mixture of fructooligosaccharide (95%) and AA (acetylated adipate distarch) added, after 2 and 24 hours of storage at 5℃, were investigated. A central composite design was used for arrangement of treatment. Different levels of fructooligosaccharide (95%), 0, 3, 6, 9, and 12%, were added to dry rice flour. In addition, different levels of AA, 0, 0.3, 0.6, 0.9, and 1.2%, were added to the same dry rice flour. The texture properties analysis using a Texture Analyzer revealed that the springiness, cohesiveness, chewiness, gumminess, adhesiveness, and hardness were significantly different. The effect of retarding retrogradation of Korean rice cakes (Karedduk) with added mixtures of fructooligosaccharide (95%) and AA showed an increasing trend as the amount of fructooligosaccharide (95%) and AA increased. Overall, the instrumental texture properties were highly correlated with the sensory characteristics. These results suggest that adding a mixture of 6% fructooligosaccharide (95%) and 1.2% AA to Korean rice cakes (Karedduk) is effective for retarding retrogradation.

The stabilizing effect of fructooligosanharide (FO) on hen's egg yolk immunoglobulin (yIgG) by heat and acid was investigated. The heat stability of yIgG at 70∼80$^{\circ}C$ was enhanced in a concentration-dependent manner by adding 0∼50% (w/v) FO to a yIgG solution. Acid-induced inactivation of yIgG was also suppressed in a concentration-dependent relationship by addition of FO. Addition of 50% FO almost completely stabilized yIgG at pH 3. The remarkable stablizing effect of FO on yIgG may enhance the use of yIgG as functional food ingredients.
