초록

Sosiho-tang, known as Sho-saiko-to in Japanese and Xiao-Chai-Hu-Tang in chinese, is an herbal medicine in Korea. It has been used for allergic reaction and chronic hepatitis disease in East Asia. This study aims to determine the anti-inflammatory effect of Sosiho-tangin lipopolysaccharide (LPS)-induced mice and Raw 264.7 cells. C57BL/6J mice were orally treated with 0.05, 0.55 and 5.55 g/kg of Sosiho-tangprior to administration of LPS. After 2 h of LPS sensitization, serum and lung tissues were obtained. Appearance of lung was confirmed by hematoxylin and eosin staining. Reverse transcription polymerase chain reaction and Western blot analysis were performed to investigate the inflammatory factors in lung tissues. In addition, the pro-inflammatory cytokines were determined in LPS-sensitized Raw 264.7 cells. Sosiho-tangameliorated the lung inflammatory responses such as alveolar hemorrhage, pulmonary alveolar wall thickening and shrinking space. Serum IL-6 level was reduced by pre-treatment of Sosiho-tang. In addition, proinflammatory cytokines including TNF-α, IL-6 and IFN-γ in lung tissue and Raw 264.7 cells were decreased in the Sosiho-tang-treated groups compared to only LPS-treated group. Furthermore, the transcription of NF-κB and phosphorylation of mitogen-activated protein kinase induced by LPS sensitization in lung tissues were significantly inhibited by pre-treatment of SSHT. In conclusion, Sosiho-tangalleviated the inflammatory TNF-α, IL-6 and IFN-γ levels, following the inhibition of NF-κB nucleation and MAP kinases phosphorylation. Additionally, the symptoms of acute liver inflammation by LPS sensitization in mice were apparently attenuated by Sosiho-tangvia suppression of pro-inflammatory cytokines and inflammatory mediators.
