In line with the ban of animal testing for cosmetics at home and abroad, the need to develop alternative test methods has increased. Traditionally, a skin irritation test was performed using rabbits, and however, now skin irritation is evaluated using a human skin model with a tissue engineering technique. The OECD has adopted Test Guideline (TG) No. 439 using four validated skin models. In Korea, the Ministry of Food and Drug Safety (MFDS) also adopted the “alternative test method guideline on cosmetics safety evaluation: skin irritation test using reconstructed human epidermis (RhE),” developed based on OECD TG 439. RhE models have various quality control criteria since there are differences in their origin and manufacturing. The quality control criteria should be met. But a skin irritation test has a limitation as it cannot classify weak irritation. The OECD is developing a new approach to address the limitation, and has recently adopted the integrated approach on testing and assessment (IATA) to fully replace animal testing. In Korea, it is necessary to establish a national strategy to develop a new approach to safety assessment following the international trend.

국내·외 화장품 동물실험 금지에 따라 국제적으로 대체시험법 개발이 활발히 이뤄지고 있다. 기존 피부자극 성은 토끼를 이용하여 평가하였으나, 최근 공학적으로 제작한 인체 피부 유사 모델을 이용하여 평가가 가능해졌다. OECD에서는 현재 TG 439에서 검증평가된 4개의 국외 모델을 인정하고 있으며, 국내는 최근에 「화장품 독성시험 동물대체시험법 가이드라인」으로 도입한 바 있다. 각 인체 피부모델은 기원 및 제작방법 등에서 차이가 있어 품질 관리 기준이 서로 다르기 때문에 이를 만족하는지 시험에 이를 반드시 포함하도록 해야 한다. 또한 동물을 이용한 피 부자극시험과 달리 약자극성을 평가하지 못하는 한계점이 있어 각 규정에 맞게 활용하는 점이 고려되어야 하며, 이 는 향후 대체시험법 활용시 항상 고려해야하는 부분으로 사료된다. 이러한 한계점을 보완하고, 동물시험을 완전 대체 하기 위해 OECD에서는 최근 통합평가접근법을 도입하고 있으며, 국내도 이러한 흐름에 대한 전략적 대응을 통해 체 계화된 안전성평가 기준이 국내 수립되도록 노력할 필요가 있다.
