Tocopherol is used as antioxidants, fragrance, skin conditioning agents and anti-evaporators in cosmetics and content is restricted to less than 20%. MFDS (Ministry of Food and Drug Safety) provides guidelines on analysis method of restricted ingredients in cosmetics for cosmetic-related industry. Because analysis method of Tocopherol for cosmetic products has not been established yet, it has been required to establish it. We used HPLC-FLD to establish analysis method of Tocopherol in cosmetics and selected Tocol as internal standard. Preconditioning method was developed to be applied to a variety of formulations such as skin, lotion and cream. In order to verify it, we validated the established method in compliance with “The guidelines for validation of the test method for medical substances”. Cross validation was conducted by testing agency and showed consistent results. Established tocopherol analysis method will be provided through “The guidelines for analysis method for the restricted ingredients in cosmetics”. It is expected that the developed method can contribute to the safety and quality control of the cosmetic products and related industry.

토코페롤은 화장품에서 산화방지제, 향료, 피부컨디셔닝제, 수분증발차단제 등의 목적으로 사용되며 20%로 한 도가 지정되어 있다. 식품의약품안전처에서는 화장품 중 배합한도 분석법 가이드라인을 통해 사용한도 성분의 시험 법을 마련하여 관련 업계에 제공하고 있으나 토코페롤은 현재 분석법이 없어 확립이 필요한 실정이다. 화장품 중 토 코페롤의 분석법을 확립하기 위해 액체크로마토그래프 형광검출기(HPLC-FLD)를 사용하였고, 내부표준물질로 토콜 을 선정하였다. 스킨, 로션 및 크림 등 다양한 화장품 제형에 대해 확립된 시험법이 적용 가능하도록 전처리법을 개 발하였다. 확립된 분석법을 검증하기 위해 ‘의약품 등 시험방법 밸리데이션에 대한 가이드라인’에 따라 밸리데이션을 진행하였다. 액체크로마토그래프를 이용한 토코페롤 분석법은 외부기관을 통해 밸리데이션 교차검증을 수행하였고, 일 관성 있는 결과를 확인하였다. 확립된 토코페롤 분석법은 향후 ‘화장품 중 배합한도 성분 분석법 가이드라인’를 통해 제공될 예정이며, 화장품의 안전 및 품질관리에 크게 기여할 것으로 사료된다.
