관광동기와 이미지가 행동의도에 미치는 영향 - 이미지변수의 매개효과를 중심으로 -

In recent literature, most of image studies involve the cognitive images while few deals with the affective images. The aim of this study is to suggest, tourism motivation develops significantly in explaining a substantial value on the visitor’s behavioral intention and how to be frequently affected on the relationships between tourism motivation and behavioral intention by cognitive images and affective images. The results of this research is presented as follows: first The tourism motivation which is composed of multi-variables; sociability, escaping daily life and relaxation, had a significant influence on the cognitive images and the affective images. second the tourism intention had a significant influence on behavioral intention and finally the cognitive images and affective images have a mediating effect partially between tourism motivation and behavioral intention.

매설가스배관 강 슬리브 보수 용접부의 피로수명에 미치는 공정변수 영향 고찰을 통한 최적공정 수립

본 연구에서는 매설가스배관의 강 슬리브 보수 용접부에 대한 피로수명에 미치는 보수 용접 공정 변수별 영향을 고찰하여 최적 공정을 수립하였다. 상용 유한요소 해석프로그램인 ABAQUS를 이용한 연계 연성 온도-응력 해석을 통해 강 슬리브 보수용접 시 보수용접부에 발생하는 용접 잔류응력을 도출하였다. 또한, 유한요소 선형탄성 응력해석을 통해 운전응력 변이도 도출하였다. 도출된 용접 잔류응력과 운전응력 변이를 입력값으로써 구조응력/파괴역학 접근법에 대입하여 강 슬리브 보수용접부의 피로수명을 평가하였다. 다양한 보수용접 공정 변수에 대한 유한요소해석과 피로평가를 수행하여 공정변수별 피로수명에 미치는 영향을 고찰하였다. 최종적으로는 고찰한 피로수명에 미치는 공정변수별 영항 결과를 토대로 공정 및 경제적 비용을 최소화하고 피로수명을 적절히 향상시킬 수 있는 최적 방안을 도출하였다.
In this study, an optimum process to improve structural integrity was established by investigating effect of the process variables on fatigue lifetime of steel-sleeve repair welds in buried gas pipeline. Residual stresses in the repair welds were derived through sequentially-coupled temperature-stress analysis using ABAQUS, which is a commercial finite element analysis program. In addition, variations of operating stresses were derived by finite element linear elastic stress analysis. Fatigue lifetimes of the steel-sleeve repair welds were evaluated by substituting the derived weld residual stresses and operating stress variations into the structural stress/fracture mechanics approach as input. Parametric study using finite element analysis and fatigue assessment for various repair welding process variables were carried out to investigate the effects of the process variables on the fatigue lifetime. Finally, based on the effects of the process variables on the fatigue lifetime, an optimal process to minimize the welding time and economic costs and to improve the fatigue lifetimes was derived.

A Fuzzy Inference based Reliability Method for Underground Gas Pipelines in the Presence of Corrosion Defects

Remaining lifetime prediction of the underground gas pipeline plays a key role in maintenance planning and public safety. One of main causes in the pipeline failure is metal corrosion. This paper deals with estimating the pipeline reliability in the presence of corrosion defects. Because a pipeline has uncertainty and variability in its operation, probabilistic approximation approaches such as first order second moment (FOSM), first order reliability method (FORM), second order reliability method (SORM), and Monte Carlo simulation (MCS) are widely employed for pipeline reliability predictions. This paper presents a fuzzy inference based reliability method (FIRM). Compared with existing methods, a distinction of our method is to incorporate a fuzzy inference into quantifying degrees of variability in corrosion defects. As metal corrosion depends on the service environment, this feature makes it easier to obtain practical predictions. Numerical experiments are conducted by using a field dataset. The result indicates that the proposed method works well and, in particular, it provides more advisory estimations of the remaining lifetime of the gas pipeline.

RRLC를 이용한 담배 연기 중 카보닐 화합물의 신속 분석

In this study, a simple gradient RRLC method for rapid determination of carbonyl compounds of cigarette smoke was developed. Within 10 min, 8 carbonyl compounds have been separated and identified on ZORBAX Eclipse XDB-C18 column ($4.6{\times}50\;mm$, $1.8\;{\mu}m$) with gradient elution using water and acetonitrile as a mobile phase. RRLC was used for the quantification of carbonyl compounds in mainstream smoke of 3R4F reference cigarette, and evaluated those efficiencies in the recovery, repeatability and reproducibility. The correlation coefficients ($r^2$) for calibration curves of carbonyl compounds were over 0.9998. The developed RRLC method was successfully applied to the analysis of smoke samples and the recoveries of carbonyl compounds were in the range of 97.5~102.1% with RSD<3.1%.

LC-MS/MS를 이용한 담배 연기 중 Heterocyclic Amines의 분석

This study was carried out to determine the analytical methods for heterocyclic amines(HAs) of the tobacco smoke by LC/MS/MS. HAs have been found in pyrolysate of protein and cooked food including protein, were known the Sugimura compound. HAs content of the smoke were known to exist very low ppb level. Especially, some of HAs are mutagenic and carcinogenic compounds. In according to IARC, the toxicity of N-heterocyclic amines classified IARC class 2A or 2B group. Precursors of these compounds are glutamic acid, protein and free amino acids including tryptophan, therefore, the precursors have been proved in cooked food continuously. This study was investigate multiple analysis methods for HAs and HAs contents of some commercial products. In this study, we used the linear type smoking machine for HAs analysis. At the ISO conditions, mainstream smoke was collected on cambridge filter pad, and then cambridge filter pad was extracted by 0.1% acetic acid. The extracted solution were passed cation exchange SPE cartridge to remove matrix, samples were analyzed using LC/MS/MS on MRM mode. From the result that optimized this methods, the correlation coefficient(R) of the individual compounds were good linearity over 0.999, recovery rate over 96% and the limit of detection were good values between 0.06 to 0.37 ng/mL, In addition, HAs content of some commercial products were in range of 0.02 to 43.8 ng/cig.

HPLC를 이용한 잎담배 중 유기산 함량 분석

This study was carried out to improve the analytical methods for determination of organic acids in tobacco leaf by HPLC. The samples for HPLC analysis were pre-treated by means of SPE. The calibration curve for each acid was linear and $R^2$ values ranged from 0.9990 to 1.0000. The limit of detection were obtained from the calibration curves and their values were between 1.39 to $4.87{\mu}g/mL$. Recovery rates of organic acids were between 88.6 % to 98.5 %. The concentrations of organic acids among the various tobacco leaves were compared to the concentration of organic acids, were in the order oriental, burley, flue-cured tobacco. In the case of flue-cured and oriental tobacco leaves, the order of concentration of organic acids was malic acid, oxalic acid, citric acid. But in the case of burley tobacco leaves, the order of concentration of organic acids was citric acid, oxalic acid, malic acid.

Solid-Phase Extraction을 이용한 담배연기 중 Aromatic Amino의 분석

A new procedure has been developed for the quantitation of aromatic amines in mainstream cigarette smoke. Two solid-phase extraction (SPE) clean up steps, using a different retention mechanisms, are required to process the samples. The first step used a cation-exchange cartridge, followed by a second step that used a cartridge with a hydrophobic retention character. The aromatic amines eluted from the second SPE cartridge are derivatized with pentafluoropropionic anhydride. This new method have advantages over other reported techniques, being sensitive, robust, and easily automated. The detection limits were ranged from 0.12 ng/mL for 1-aminonaphthalene to 0.16 ng/mL for 3-aminobiphenyl and the recoveries were from 97 to 106%. Compared with other reports for analysis of 2R4F reference cigarette, this method shows a close analytical data and good repeatability.

Background: To develop a novel treatment strategy for hepatitis B virus infection, a major cause of liver chirosis and cancer, we aimed to make human monoclonal antibodies inhibiting RNase H activity of P protein playing in important role in HBV replication. In this regard, phage display technology was employed and demonstrated as an efficient cloning method for human monoclonal antibody. So this study analysed the usability of human monoclonal antibody as protein based gene therapy. Methods: RNase H of HBV was expressed as fusion protein with maltose binding protein and purified with amylose resin column. Single chain Fv (scFv) phage antibody library was constructed by PCR cloning using total RNAs of PBMC from 50 healthy volunteers. Binders to RNase H were selected with BIAcore 2000 from the constructed library, and purified as soluble antibody fragment. The affinity and sequences of selected antibody fragments were analyzed with BIAcore and ABI automatic sequencer, respectively. And finally RNase H activity inhibiting assay was carried out. Results: Recombinant RNase H expressed in E. coli exhibited an proper enzyme activity. Naive library of $4.46{\times}10^9cfu$ was screened by BIAcore 2000. Two clones, RN41 and RN56, showed affinity of $4.5{\times}10^{-7}M$ and $1.9{\times}10^{-7}M$, respectively. But RNase H inhibiting activity of RN41 was higher than that of RN56. Conclusion: We cloned human monoclonal antibodies inhibiting RNase H activity of P protein of HBV. These antibodies can be expected to be a good candidate for protein-based antiviral therapy by preventing a replication of HBV if they can be expressed intracellularly in HBV-infected hepatocytes.
