Our continuing interest on Garcinia and Mesua species has led us to carry out a detail study on the chemistry of the root bark of Garcinia mangostana (Guttiferae) since this part of the plant has not been investigated before, and the strm bark of Mesua corneri (Guttiferae) an uninvestigated species. This study has yielded six xanthones, ${\alpha}-mangostin$ (1), ${\beta}-mangostin$ (2), ${\gamma}-mangostin$ (3), garcinone-D (4), mangostanol (5) and gartanin (6) from Garcinia mangostana and two xanthones rubraxanthone (7) and inophyllin B (8) from Mesua corneri. Structural elucidations were achieved using $^1H,\;^{13}C$ NMR and MS data. The crude hexane and chloroform extracts of the root bark of Garcinia mangostana and the hexane extract of the stem bark of Mesua corneri were found to be active against CEM-SS cell lines with $IC_{50}$ values less than $30\;{mu}g/ml$. Moreover, ${\gamma}-mangostin$ gave a very low $LC_{50}$ value of $4.7\;{mu}g/ml$ while rubraxanthone gave an $LC_{50}$ value of $5.0\;{mu}g/ml$ indicating these two compounds to be potential lead compounds for anti-cancer activity against the CEM-SS cell line. This paper reports the isolation and identification of these compounds as well as bioassay data for the crude extracts, ${\gamma}-mangostin$ and rubraxanthone.

Fracture toughness defined near the initiation of stable crack growth is investigated by R-curve and Direct Current electric Potential Determination(DCPD) under mode I plane strain conditions for CT specimen with 25.4mm thickness of SS400 steel. Fracture toughness. $J_{IC}$lit near crack tip of CT specimen by R-curve is 17.14 $kg_{f}/mm$ and however. its value by DCPD is 22.82 $Rg_{f} mm$ The value of fracture toughness by DCPD is larger than that by R-curve. Therefore, it is suggested that the evaluation of fracture toughness by R-curve is optimum than by DCPD, when considering amount of crack growth about each of fracture toughness.

...ssay로 실험한 결과, 3종의 암세포주(HepG2, HeLa, MCF-7) 모두 돌나물의 ethylether 분획층(SSMEE)에서 아주 높은 암세포 증식억제 효과를 보였으며, ethylacetate 분획층(SSMEA)에서도 유의적인 암세포 증식억제 효과를 나타내었다. 또한, 돌나물 분획물의 암예방 QR유도 활성을 HepG2 세포주를 이용하여 실험한 결과, 다른 분획층에 비해 비극성 용매층인 ethylether 분획층(SSMEE)과 hexane 분획층(SSMH)에서 유의적으로 QR유도 활성을 증가시키는 것으로 나타났다. 한편, 돌나물과 도라지와의 일정비 조합을 통해 상승효과를 본 결과, $IC_{50}$/이 되는 도라지의 butanol층 (PGMB)dfm 일정량 첨가했을 때 용매별 돌나물의 분획층 모두에서 암세포 증식억제를 상승시키는 효과를 나타내었고, 암예방 QR 유도활성 효과도 훨씬 상승하였으므로, 돌나물 조리시 유사한 부재료를 사용하는 도라지의 성분을 일정량 첨가하여 겉절이 무침이나 김치로 이용될 때 돌나물 성분의 생리활성 효과가 더욱 상승될 것으로 사료되며, 이후 돌나물의 생리활성 물질 구조 및 기전 규명에 유익한 자료가 될 것으로 사료된다.

Anticarcinogen is one of the major strategies for cancer control. It is well established that dietary factors play an important role in modulating the development of certain types of human cancer. We investiagted the anticarcinogenic effects of Sedum sarmentosum Bunge (SS) with Platycodon grandiflorum A. extracts on HepG2, HeLa and MCF-7 cell lines. By the MTT assay, among the five partition layers of methanol extract of SS (SSM), the ethylether partition layer of SS (SSMEE) showed the strongest cytotoxic effects on all cell lines. We also investigated the synergistic effect of the combination of SS and PG extracts on growth inhibition of the HepG2, HeLa and MCF-7 cell lines compared to the effects of five partition layers of SSM. Combination of SS and PG extracts significantly increased cytotoxic effects on all cell lines. Therefore, we were able to conclude that ethylether partition layer, SSMEE might have potentially useful cytotoxic materials on all the human cancer cells which we used. And we could suggest that the combination of SS with PG enhanced the anticarcinogenic effect on HepG2, HeLa and MCF-7 cell lines. We also determined QR activity of partition layers of SSM, among them, SSMEE on HepG2 cells showed the highest QR activity, 3.21 as control value of 1.0.

Objectives. Sodium salicylate (SS) is well known for its ototoxic properties that induce functional and morphological changes in the cochlea and brain. Ginkgo biloba extract (GBE) has been widely used for treatment of various neurodegenerative diseases; however, its effects on salicylate-induced ototoxicity remain unclear. Herein, we examined the effects of EGb 761 (EGb), a standard form of GBE, on the plasticity of the N-methyl-D-aspartate receptor subunit 2B (GluN2B) in the inferior colliculus (IC) following SS administration. Methods. Seven-week-old Sprague Dawley rats (n=24) were randomly allocated to control, SS, EGb, and EGb+SS groups. The SS group received a single intraperitoneal SS injection (350 mg/kg), the EGb group received EGb orally for 5 consecutive days (40 mg/kg), and the EGb+SS group received EGb for 5 consecutive days, followed by an SS injection. The auditory brainstem responses (ABRs) were assessed at baseline and 2 hours after SS administration. GluN2B expression was examined by Western blot and immunohistochemistry. Results. There were no significant differences in ABR threshold shifts among the groups. The expression of the GluN2B protein normalized by which of glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was significantly lower in the EGb+SS group, as compared to the SS group (P=0.012). Weak and diffused GluN2B immunoreactivity was detected in the IC neural cells of the EGb+SS group, while those of the SS group exhibited strong and diffused GluN2B positivity. Conclusion. EGb may play a role in regulating the GluN2B expression in the IC of salicylate-induced ototoxicity model.

The objective of this study was to determine the influence of quality grade (QG) on meat tenderness characteristics of ten major muscles from Hanwoo steers. A total of 25 Hanwoo carcasses ($5\;carcasses{\times}5\;QGs$) were selected. Intramuscular fat content, collagen content, sarcomere length, and Warner-Bratzler shear force (WBSF) of Longissimus thoracis (LT), Longissimus lumborum (LL), Psoas major (PM), Semisponals (SS), Triceps brachii (TB), Semimembranosus (SM), Gluteus medius (GM), Rectus Abdominis (RA), Superficialis flexor (SF), and Internal and external intercostal (IC) were determined. IC had the highest fat content, followed by LT, RA, LL, PM, GM, SS, SF, TB, and SM. High-fat muscles such as LT, LL, IC, RA, and PM had significantly (p<0.05) different fat contents among QGs. Collagen contents were significantly (p<0.05) different among QGs. With decreasing QG, increasing collagen content was found in muscles. There were significant (p<0.05) differences in sarcomere length among QGs of several muscles. However, no significant (p>0.05) difference in sarcomere length was found among QGs for LL, PM, or RA muscle. PM had the lowest WBSF, followed by LL, LT, RA, IC, GM, SM, SF, SS, and TB. WBSF of QG $1^{{+}{+}}$ was lower than that of QG 1 for SS, TB, and SM. All muscles of QG 1 showed lower WBSF than QG 3 except TB or IC. Results of this study suggested that differences in WBSF among these 10 muscles by QG were due to differences in collagen content and sarcomere length.

In drug discovery or preclinical stages of development, potency parameters such as $IC_{50}$, $K_i$, or $K_d$ in vitro have been routinely used to predict the parameters of efficacious exposure (AUC, $C_{min}$, etc.) in humans. However, to our knowledge, the fundamental assumption that the potency in vitro is correlated with the efficacious concentration in vivo in humans has not been investigated extensively. Thus, the present review examined this assumption by comparing a wide range of published pharmacokinetic (PK) and potency data. If the drug potency in vitro and its in vivo effectiveness in humans are well correlated, the steady-state average unbound concentrations in humans [$C_{u_-ss.avg}=f_u{\cdot}F{\cdot}Dose/(CL{\cdot}{\tau})=f_u{\cdot}AUCss/{\tau}$] after treatment with approved dosage regimens should be higher than, or at least comparable to, the potency parameters assessed in vitro. We reviewed the ratios of $C_{u_-ss.avg}$/potency in vitro for a total of 54 drug entities (13 major therapeutic classes) using the dosage, PK, and in vitro potency reported in the published literature. For 54 drugs, the $C_{u_-ss.avg}$/in vitro potency ratios were < 1 for 38 (69%) and < 0.1 for 22 (34%) drugs. When the ratios were plotted against $f_u$ (unbound fraction), "ratio < 1" was predominant for drugs with high protein binding (90% of drugs with $f_u{\leq}5%$; i.e., 28 of 31 drugs). Thus, predicting the in vivo efficacious unbound concentrations in humans using only in vitro potency data and $f_u$ should be avoided, especially for molecules with high protein binding.

This report compares the selective cytotoxic effects of Doenjang fermented by various Bacillus strains (Bacillus sp. SS9, SSA3, and PM3) on human liver cell lines with that of conventional Doenjang (DTY, DTG, and DTK) and commercial Doenjang (DCM, DCD, and DCS). To investigate selective cytotoxic effects of Doenjang extracts, the cell density of HepG2 (Hepatocellular carcinoma) and CCL-13 (cells derived from human normal liver) was estimated after addition of the extracts by using a viable cell counting method. The maximum selectivity ratio ($IC_{50}$value against CCL-13/$IC_{50}$ value aganist HepG2) was observed by PM3 (extracts of Doenjang fermented with Bacillus sp. PM3). As for morphological changes shown by the addition of PM3 into HepG2 and CCL-13 cultures, HepG2 was significantly disrupted, however, CCL-13 was not affected. Also, the growth rate of HepG2 was decreased significantly by the addition of PM3. Consequently, PM3 showed a more detrimental effect on HepG2 than that on CCL-13.

Three film deposition systems (pulsed laser deposition, sputtering, and evaporation) equipped with reel-to-reel(R2R) metal tape moving apparatus were installed and used to deposit multi-layer oxide thin films. Both RABiTS and IBAD texture templates are used. IBAD template consists of CeO$_2$(PLD)/YSZ(IBAD) on stainless steel(SS) metal tape, and RABiTS template has the structure of CeO$_2$/YSZ/Y$_2$O$_3$ which was continuously deposited on Ni-alloy tape using R$_2$R evaporation and DC reactive sputtering in a deposition system designed to do both processes. 0.4 m-long coated conductor with Ic(77 K) of 34 A/cm was fabricated using RABiTS template. 0.5 m and 1.1 m-long coated conductor with Ic(77 K) of 41 A/cm and 26 A/cm were fabricated using IBAD template.

The main purpose of this study was to recycle the used oxide slurry using filters as a new method. As a result, Ultra Fine(UF) Filter could distinguish silica from the used oxide slurry and Reverse Osmosis(RO) Filter could distinguish Deionized(DI) Water and chemistry from chemistry solution. The tetraethylorthosilicate removal rate was almost the same as the number of recycle polishing was increased, when it was modified by slightly adding new SS-12 slurry. The microscratch didnt found as the number of recycle polishing was increased.
