Carnosine is an endogenously synthesized histidine-containing dipeptide which is present in brain and other various organs in the body.1,2) Carnosine has been known to delay aging and rejuvenate senescent cultured human fibroblasts. Several studies suggest that carnosine readily reacts with aldehydes and ketones and consequently protects macromolecules against their cross-linking actions. The present study revealed that the carnosine suppressed levodopa or excess cytosolic free dopamine (DA)-induced quinone generation in the striatum of parkisonism models and in dopaminergic cells. Furthermore, carnosine stabilized free DA and DA quinone as dopaminergic neuron-specific oxidative stress by rapid conversion to stable melanin. The stabilizing effect of carnosine against cytotoxic DA quinones may play an important role in its neuroprotective machanism in the brain.
