Identification of the chemicals that pose a hazard to the developing nervous system is the first step in reducing human exposure and preventing health problems. In response to the need for more efficient methods to identify potential developmental neurotoxicants, we designed a cell based chip to detect the environmental chemical effects on neurite outgrowth in PC-12 cell, which is a cancer cell line derived from a pheochromocytoma of the rat adrenal medulla. Cell based chip were fabricated by binding synthetic oligopeptide (RGD) onto a gold electrode. PC- 12 cells were immobilized on the gold electrode. Induction of the neurite cell to grow was observed by the neurite growth factor (NGF). Electrochemical behavior of the effect of the environmental chemical (aldrin) on the neurite outgrowth was measured using a cyclic voltammogram. Voltammetric current peak behavior shows that as the concentration of aldrin increases, the measured peak current decreases. This indicates a decrease in cell viability. The cell chip used in this study can be applied to detect the inhibitory effects of a large number of environmental toxins on neurite outgrowth.
