The aim of this study is to evaluate the sensitivity and specificity of the DNA chip developed in previous study by comparing conventional clinical diagnostic methods and to verify the sensitivity and simplicity of the DNA chip according to clinical DNA sources. Two hundred twenty seven participants volunteered for this test. Diagnosis was first performed by a slit-lamp eye examination, which is a common medical diagnostic method, and then each DNA sample from peripheral blood was analyzed by DNA chip and sequencing simultaneously. All the tests were performed independently as blind tests. The evaluation of slit lamp examination and DNA chip was finally done by comparing the DNA sequencing results. Seven heterozygous GCD II patients were selected to examine how DNA chip is sensitive and which samples are applicable except for blood. DNA were extracted from their buccal epithelial cells, hair roots, epidermis stuck on skin patch sticker and blood. A total of 28DNA samples were collected and sent for further analysis by DNA chip and sequencing. DNA sequencing showed that of the 227 participants, 124 are GCD II patients (54.6%) and 103 are normal healthy individuals - non GCD II (45.4%). The sensitivity and specificity of DNA chip were both 100%, while the sensitivity and specificity of slit lamp method were 99.19% and 100%, respectively. When DNA samples from different clinical sources of 7 patients were tested usingDNA chip and sequencing simultaneously, only DNA chip was able to give the results directly in all types of samples, but DNA sequencing was only successful using DNA from blood. It was firmly concluded that DNA chip allows rapid and successful diagnosis of GCD II, which is better than conventional clinical method and it also allows simple and easy detection with various types of clinical samples better than DNA sequencing.
