Preparation of a Mep45 Protein-fused Substrate for Phophorylation Assay to Detect Protein Kinase C using Radioisotope Detection Technique for the Development of a Biochip
Basic biological and medical research continues to provide the building blocks necessary to develop solutions to human health problems and to preserve and repair the natural environment. In this study, we investigated the feasibility of applying a RI detection technique to a protein kinase C assay for the development of a diagnostic biochip. Using the mep45 gene encoding the 45 kDa major envelope protein (Mep45) of Selenomonas ruminantium, a Mep45-fused substrate was cloned and purified for use as substrate to detect the activity of protein kinase C. The resultant MFS-PKC protein was used as a substrate for a protein kinase C assay and the signal intensity of the kinase assay system was examined with varying concentrations of the kinase. The results are found to be promising and can be applied when using a RI detection technique for developing a highly sensitive biochip to determine the enzyme activity for a disease diagnosis and subsequent new drug development.
