Thraustochytrium aureum ATCC 34304 strain produce about 35-40% DHA (docosahexaenoic acid) in lipids by typical FAS (fatty acids synthesis) pathway. For research of FAS pathway from T. aureum ATCC 34304, it has problems with being several over-sized chromosomal DNA and being mitochondrial DNA. An electrophoretic karyotype of Thraustochytrium aureum has been separated by pulsed-field gel electrophoresis. the T. aureum chromosome size was estimated by using the Saccharomyces cerevisae as size standard. Totally we identified as 13 chromosomal band and its whole genome size is estimated to be approximately 12.9 Mbp. The seperation of chromosome by electrophoretic technique is very useful for studying the genetic and molecular findings. Also this results of number and size of the chromosome are very helpful for the construction of chromosome-specific libraries and gene interest in physical map linkage.
