Polyunsaturated fatty acids (PuFAs) like EPA and DHA have attracted public attention because of their beneficial effects in human health. To date, several genes encoding critical enzymes like desaturases and elongases involved in the biosynthesis of PuFAs have been cloned from various organisms. A TaELO elongase gene was isolated from Thraustochytrium aureum ATCC 34304 and cloned into Pichia pastoris GS115 using pPIC3.5 vector. C18-20 elongase and C20-22 elongase was expressed through multistep in P. pastoris GS115. The temperature is an important parameter for optimization of elongase expression in yeast systems. In this report, we demonstrated the effects of cultivation temperature on TaELO elongase gene expression in C18-20 elongation step.
