The Efficacy of Polynucleotide and Hyaluronic Acid Combination Therapy in an Animal Model of Induced Osteoarthritis

This study investigates the effects of a combination of Polynucleotide (PN) and Hyaluronic Acid (HA) on osteoarthritis (OA) by administering the compound for four weeks. Osteoarthritis was induced in male Sprague-Dawley rats using monosodium iodoacetate (MIA), and PNHA was administered intra-articularly at a dose of 50 μL once a week for four weeks. Our study included assessments of mortality, general symptoms, body weight changes, gross findings at necropsy, and histopathological examinations.The results demonstrated that PNHA significantly alleviated OA symptoms, particularly in promoting cartilage regeneration and enhancing joint function. No deaths or severe adverse effects were observed in the PNHA group, and the body weight changes were similar to those in the control group. Moreover, the weight-bearing index significantly improved in the PNHA-treated group, indicating enhanced joint function. Histopathological analysis revealed cartilage regeneration and structural improvement in the joint tissue of the PNHA group. Unlike prior studies that focused on single-agent therapies, this study provides integrated preclinical evidence of the combined PNHA therapy’s synergistic effects in an OA model. These results suggest that PNHA is an effective treatment for alleviating OA symptoms and promoting cartilage regeneration. Additionally, We need further clinical studies to solidify the safety and efficacy of the Polynucleotide and Hyaluronic Acid combination therapy.

본 연구는 폴리뉴클레오티드(PN)와 히알루론산(HA) 복합제가 골관절염(OA)에 미치는 효능을 4주 동안 평가하였다. 수컷 Sprague-Dawley 랫드에서 monosodium iodoacetate (MIA)로 OA를 유도한 뒤, PNHA를 50 μL씩 주 1회, 총 4주간 관절강내 투여하였다. 사망률, 일반 증상, 체중 변화, 부검 시 육안 소견 및 조직병리학적 변화를 분석하였다. 그 결과 PNHA 투여군은 대조군에 비해 OA 증상이 유의하게 감소하였으며, 특히 연골 재생 촉진과 관절 기능 개선 효과가 두드러졌다. 투여 기간 동안 사망이나 중대한 이상반응은 관찰되지 않았고, 체중 변화 또한 대조군과 유사하였다. 하중 지수(weight-bearing index)는 PNHA 투여군에서 유의하게 증가하여 관절 기능 향상을 시사하였다. 조직병리학적 분석에서도 PNHA 투여군의 관절 조직에서 연골 재생 및 구조적 개선이 확인되었다. 기존 연구들이 단일 성분의 효과에 주목한 데 반해, 본 연구는 PN과 HA의 복합 투여가 갖는 시너지 효과를 MIA 유도 OA 모델에서 통합적으로 입증한 것이 특징이다. 이러한 결과는 PNHA 병용 요법이 OA 증상 을 완화하고 연골 재생을 촉진하는 효과적인 치료 전략이 될 수 있음을 시사한다. 다만, PNHA의 안전성과 유효 성을 확립하기 위해 추가적인 임상 연구가 필요하다.
