Change of Coatomer Subunit Gamma-2 Protein during Estrous Cycle in Porcine Endometrium

This study investigated change of protein trafficking-related coatomer subunit gamma- 2 (COPG2) using proteomics analysis during estrous cycle in porcine endometrium. The endometrium tissues were collected from porcine uterus, were classified according to ovary morphology before or after ovulation. The COPG2 protein was found by two dimensional electrophoresis and MALDI-TOF/MS-MS, and COPG2 protein expression and location were observed using western blotting and immunofluorescence in proliferation and secretion phase endometrium tissues. Porcine endometrial epithelial cells (PEECs) were isolated from endometrium, and influence of progesterone (P4) on COPG2 protein change was observed using western blotting, flow cytometry and immunocytochemistry in PEECs. In result, eighteen protein spots were changed between proliferation and secretion phase endometrium, and molecular functional analysis demonstrated that one of theses, COPG2, was involved in protein trafficking between endoplasmic reticulum and Golgi apparatus. Additionally, the COPG2 was increased in tissue and PEECs from secretion phase compared to proliferation phase. The COPG2 protein was significantly increased by 40 ng/mL P4 treatment group compared to 0 and 10 ng/mL treatment groups P4 in PEECs. These results may provide understanding of protein trafficking in endomerium during estrous cycle, and will be used in basic study data for relation of P4 and protein trafficking in female reproductive tracts.
