As a brown algae, Sargassum sp. are found throughout tropical and subtropical areas of the world and are valuable source to produce various bioactive secondary metabolites. In this study, effect of Sargassum sp. on adipocyte differentiation in 3T3-L1 adipocytes was investigated using methanol extracts from Sargassum horneri, Sargassum thunbergii and Sargassum hemiphyllum. The level of lipid accumulation was determined by measuring Oil-Red O staining at the end of differentiation. In comparing analysis, methanol extract of S. thunbergii indicated the lowest lipid accumulation in a dose-dependent manner. Lipid accumulation along with the expression of several genes associated with adipogenesis was examined at the end of differentiation. The expression levels of adipose genes were examined using reversetranscription polymerase chain reaction (RT-PCR) analysis. S. thunbergii extract showed significantly reduced lipid accumulation and downregulated peroxisome proliferator-activated receptor-γ and CCAAT/ enhancer-binding proteins α in a dose-dependent manner. Therefore, S. thunbergii extract could be developed as a functional ingredient effective in improving obesity.
