Streptomyces strains are best known for their ability to produce important secondary metabolites. Also, two-component system performs an important role in secondary metabolite production of actinomycetes. Therefore, the two-component system was cloned from S. acidiscabies and its function has been studied. Two Genes (tcsK and tcsR) encoding the sensor kinase (TCSK) and the response regulator (TCSR) were searched by PCR, respectively, and then intact 1.6-kb tcsK and 0.7-kb tcsR were obtained from S. acidiscabies genome by fosmid library construction and PCR screening. To identify the function of the cloned genes, the high expression vectors were constructed by using pSET152ET, and then introduced into S. lividans TK24 and S. acidiscabies. It was determined that the high expression of tcsK and tcsR induced 5.4-fold and 5.2-fold increases in actinorhodin production of S. lividans TK24, respectively. Also, tcsK disruption in S. acidiscabies caused change in morphological differentiation and decreased WS5995B production, but did not affect thaxtomin production.
