In this study, anti-inflammatory effects of crude extracts from Ecklonia cava and their solvent fractions were evaluated using several different activity tests, including degree of occurrence of intracellular reactive oxygen species (ROS), nitric oxide (NO), mRNA expressions of cyclooxygenase-2 (COX-2), and inducible nitric oxide synthase (iNOS) in LPS-activated mouse macrophage Raw 264.7 cells. The levels of intracellular ROS were measured using 2',7'-dichlorofluorescin diacetate (DCF-DA) as fluorescence probe. The generation of NO products was determined based on the Griess reaction. All of them not only exhibited the significant scavenging activity on the generation of ROS but also inhibited LPS-induced NO production without a notable cytotoxicity in Raw 264.7 cells. In addition, they suppressed the mRNA expression of iNOS and COX-2 in LPS-activated Raw 264.7 cells. Among them, n-buthanol fraction, having high polyphenol content, exhibited the strongest anti-inflammatory effects. Therefore, these results suggest that novel anti-inflammatory compounds can be isolated from E. cava.
