We investigated whether visfatin gene expression, upregulated by hypoxia in cancer cells1), was associated with differentiation of neuronal cells. RT-PCR analysis, morphological observations, and immunostaining revealed that CoCl2, a hypoxic mimetic agent, at 75 microM increased expression of the visfatin gene, and induced neurite outgrowth in PC-12 rat pheochromocytoma cells2). Visfatin not only induced neurite outgrowth but also significantly upregulated Vigin (visfatin - inducible gene in neuritogenesis) mRNA levels. Therefore, considering that vigin has been known to be essential for neurite outgrowth and neuroprotective effect, our present results suggest that visfatin-mediated vigin signaling pathway may play important roles in neuritogenesis and neuroprotection. 1) Bae SK, Kim SR, et. al.
