Acanthoside-D, contained in Acanthopanax chilsanensis is known as a ginseng-like substance. This work was focused to set up analytical and preparative conditions for Acanthoside-D purification. The ethanol extract from the powder of the trunk of Acanthopanax chilsanensis was partitioned with hexane. A μ-Bondapak C18 (3.9×300 mm, 10 μm) column was used to separate Acanthoside-D from the trunk of Acanthopanax chilsanensis. From the experimental results, the
mobile phase used for isolating Acanthoside-D from the extract was water/acetonitrile/methanol=80/14/6 %(v/v). The flow rate of mobile phase was 1.0 mL/min, and UV wavelength was fixed at 210 nm. Finally on a semi-preparative column (3.9×300 mm, 15 μm, Lichrospher 100RP-18) with the same mobile phase composition, the allowable maximum
injection volume increased to 250 ㎕.

지리산 오갈피에 주로 포함된 Acanthoside-D은 인삼과 유사한 물질로 알려져 있다. Acanthoside-D에 대한 분석용뿐만 아니라 제조용에서의 실험조건을 구하는 것이 연구의 목적이 된다. 지리산 오갈피의 줄기의 파우더를 ethanol 용액으로 추출한 후 hexane으로 분배하였다. 정제된 추출액을 μ-Bondapak
C18 (3.9×300 mm, 10 μm) 컬럼을 이용하여 지리산 오갈피 줄기에 포함된 Acanthoside-D를 분리하기 위한 분석조건을 확립하였다. 본 연구의 결과에 의하면 목적물질인 Acanthoside-D를 분리하기 위해 이동상의 조성은 water/ acetonitrile/methanol=80/14/6 %(v/v)이었고, 유속은 1.0 mL/min, UV 검지기는
210 nm로 고정하였다. 위의 분석조건을 이용하여 반제조용 컬럼 (3.9×300 mm, 15 μm, Lichrospher 100RP-18)에서 분리가능한 최대 주입량은 250 μL이었다.
