Minocycline is a semisynthetic second-generation tetracycline that can easily penetrate the blood-brain barrier. Its neuroprotective properties were demonstrated in various disease models such as cerebral ischemia, amyotrophic lateral sclerosis, and Huntington's disease. In addition, minocycline was found to be able to block 6-OHDA- and MPTP-induced dopaminergic neurodegeneration associated with Parkinson's disease (PD). The present study investigated neuroprotective properties of minocycline in a paraquat-induced mouse model of PD. Paraquat, a quaternary nitrogen herbicide, induced the c-Jun N-terminal kinase (JNK) and caspase-3 activation in Substantia Nigra pars compacta (SNpc) dopaminergic neurons in vivo. These effects were attenuated by minocycline (40mg/kg, i.p.) pretreatment. Results in this study support a role of minocycline against toxic insults and suggest that its therapeutic potential for PD
