α-Lipoic acid is a widely occurring coenzyme found in most prokaryotic and eukaryotic microorganisms1), as well as many plant and animal tissues2). Although α-lipoic acid has been widely and successfully used as an orally administered prophylactic and therapy for a variety of diseases including heart disease3), metabolic engineering has never been applied for α-lipoic acid production to date. Here we present a preliminary study of metabolic engineering for the efficient lipoic acid production. Three genes encoding lipoate synthase, lipoyl transferase, and lipoate-protein ligase were identified via sequence analysis of the whole genome of Pseudomonas fluorescens, and subsequently cloned and transformed into α -lipoic acid producing E. coli. The resulting recombinant, E. coli LipABL showed significantly higher α-lipoic acid production and content than the wild type E. coli. This work describes the first metabolic engineering study with the enzymes involved in α-lipoic acid biosynthesis.
