Metabolic profiles of Wolfiporia cocos (Polyporaceae, Basidiomycota) mycelia cultivated under light and dark condition

Wolfiporia cocos (F.A. Wolf) Ryvarden & Gilb. (syn., Poria cocos F.A. Wolf), a basidiomycetous fungus having subterranean growth habit, usually forms compact and hard masses of mycelium, called sclerotium, to overcome stressful environmental conditions. In East Asian countries, it has been regarded a main material to compound traditional medicines for invigorating vital energy. In the traditional cultivation of W. cocos, the inoculated pine logs were buried into the soil. Recently, a new shelf cultivation technique using plastic bag packed with pine sawdust and rice bran was developed. In the present study, the alternation of metabolic profiles in W. cocos mycelia cultivated under light and dark conditions was investigated using gas chromatography mass spectrometry (GC-MS) and high performance liquid chromatography (HPLC). We used a commercial strain of W. cocos, Bokryeong 1-ho, preserved in the National Institute of Horticultural and Herbal Science. The GC-MS data showed that the contents of sugars (e.g., trehalose, maltose, and xylitol), proline, and glycerylmonostearate remarkably increase in the light condition, while those of amino acids (e.g., glycine and cystathionine), organic acids (e.g., malic acid and fumaric acid), and some nucleosides increased in the dark condition. Pachymic acid is a natural triterpenoid having antiviral, antitumor and anti-inflammatory properties. Our quantitative estimation of pachymic acid in the both mycelial samples represented that more pachymic acid are produced under the light condition with statistical significance. * These authors contributed equally to this work.
